Morphological Expression of KIT Positive Interstitial Cells of Cajal in Human Bladder

Abstract: PurposeWe investigated the 3-dimensional morphological arrangement of KIT positive interstitial cells of Cajal in the human bladder and explored their structural interactions with neighboring cells.Materials and MethodsHuman bladder biopsy samples were prepared for immunohistochemistry/confocal or transmission electron microscopy.ResultsWhole mount, flat sheet preparations labeled with anti-KIT (Merck, Darmstadt, Germany) contained several immunopositive interstitial cell of Cajal populations. A network of ste… Show more

“…We and others recently demonstrated that ICC markers are expressed in BICCs, including c-kit, anoctamin 1, gap junction protein connexin 43, and CD34 (88). These cells are closely associated with intramural nerves and BSM cells (89)(90)(91). The overlying schematic in Figure 9 illustrates the position they occupy in the bladder wall.…”

“…They suggested that existence and survival of ICC mainly rely on neural release of trophic factors. [12] A recent study by Johnston et al [6] showed that the human bladder contains a network of ICCs in the lamina propria, which is connected to cholinergic nerve plexus. Perivascular ICCs are localized close to the vascular smooth muscle cells, whereas the detrusor ICCs are observed in smooth muscle bundles and associated with nerves, controlling the bladder contractility.…”

“…Perivascular ICCs are localized close to the vascular smooth muscle cells, whereas the detrusor ICCs are observed in smooth muscle bundles and associated with nerves, controlling the bladder contractility. [6] Piaseczna-Piotrowska et al [13] had examined the distribution of ICCs in different parts of the overactive neurogenic bladder wall of the children with MMC. They had found that the ICC are widely located in the trigone than the anterior wall of the corpus of the urinary bladder.…”

“…It is known that c-kit is a specific marker for ICs [4,8,12]. Additionally, antibodies targeting connexin 43 [8,14] and vimentin [8,15] molecules were also used to stain ICs in the human urinary tract. Therefore, we used antibodies targeting c-kit, connexin 43 and vimentin in order to stain and detect ICs in the human urinary tract tissues.…”
