DNA barcoding without assessing reliability and validity causes taxonomic errors of species identification, which is responsible for disruptions of their conservation and aquaculture industry. Although DNA barcoding facilitates molecular identification and phylogenetic analysis of species, its availability in clariid catfish lineage remains uncertain. In this study, DNA barcoding was developed and validated for clariid catfish. 2,970 barcode sequences from mitochondrial cytochrome c oxidase I (COI) and cytochrome b (Cytb) genes and D-loop sequences were analyzed for 37 clariid catfish species. The highest intraspecific nearest neighbor distances were 85.47%, 98.03%, and 89.10% for COI, Cytb, and D-loop sequences, respectively. This suggests that the Cytb gene is the most appropriate for identifying clariid catfish and can serve as a standard region for DNA barcoding. A positive barcoding gap between interspecific and intraspecific sequence divergence was observed in the Cytb dataset but not in the COI and D-loop datasets. Intraspecific variation was typically less than 4.4%, whereas interspecific variation was generally more than 66.9%. However, a species complex was detected in walking catfish and significant intraspecific sequence divergence was observed in North African catfish. These findings suggest the need to focus on developing a DNA barcoding system for classifying clariid catfish properly and to validate its efficacy for a wider range of clariid catfish. With an enriched database of multiple sequences from a target species and its genus, species identification can be more accurate and biodiversity assessment of the species can be facilitated.

Ikan lele Mutiara merupakan salah satu strain unggul hasil pemuliaan di Indonesia. Tujuan dari penelitian ini adalah untuk mempelajari status genetik ikan lele Mutiara (Clarias sp.) dari variasi sekuen parsial gen COI dari mitokondria. Ikan uji yang digunakan berupa 15 ekor ikan lele Mutiara. DNA genom ikan lele Mutiara diekstraksi dari sirip ekor, kemudian fragmen DNA diamplifikasi dengan primer spesifik COI dan selanjutnya dilakukan sekuensing. Hasil sekuens yang diperoleh kemudian dianalisis estimasi substitusi nukleotida, pohon filogenik, estimasi jarak genetik, dan komposisi asam amino residu dengan membandingkan data hasil sekuens dari ikan lele C. macrocephalus, C. batrachus, C. fuscus. dan C. gariepinus yang tersedia di database GenBank. Hasil analisis pohon filogenetik menunjukkan bahwa ikan lele Mutiara terpisah dengan kelompok ikan lele yang lain. Parameter lainnya memperkuat analisis sebelumnya bahwa ikan lele Mutiara tidak hanya memiliki perbedaan genetis dengan spesies ikan lele lainnya, tetapi juga dengan jenis ikan lele Afrika (C. gariepinus).Mutiara catfish is one of the superior strains resulting from breeding program in Indonesia. This study aims to assess the genetic status of the Mutiara catfish (Clarias sp.) by looking at the diversity in the partial sequence of mitocondrial COI gene and to add genetic data of economically important freshwater fish in Indonesia. The sample fish used were a collection of 15 Mutiara catfish. Data analysis consisted of estimation of nucleotide substitution, phylogenetic tree, estimation of genetic distance, and amino acid composition by comparing secondary data from sequences of C. macrocephalus, C. batrachus, C. fuscus. and C. gariepinus wich was available at Genbank NCBI. The results of the phylogenetic tree analysis showed that Mutiara catfish were separated from other catfish groups. Other parameters also show that Mutiara catfish not only have genetic differences with other Clarias species, but also with C. gariepinus there are also genetic differences.

Feeding is an important aspect of fish farming activities and could determine the quality of fish. African catfish (Clarias gariepinus) is an omnivorous freshwater fish that is high in protein and economical value. Catfish nursery generally uses natural feed and artificial feed to ensure the diet requirements fullfiled. Good feeding, in quality or quantity, is an important aspect to produce a high quality breed. A good feed is the basis of improving cultured catfish growth and survival performance. The purpose of this article is to review types of feed, feeding practices, and their enriched alternatives for catfish rearing.
