An efficient and reproducible in vitro plant regeneration system from shoot apices was developed in Jatropha curcas. Benzylaminopurine (BAP; 2.5 µM) was most effective in inducing an average of 6.2 shoots per shoot apex. Incorporation of gibberellic acid (GA 3 ; 0.5 µM) to basal medium was found essential for elongation of shoots. The BAP-habituated mother explants continuously produced shoots during successive subculture without any loss of morphogenic potential. The shoots rooted efficiently on half-strength MS medium. The rooted plantlets were acclimatized with more than 98 % success and the plants transferred to soil:compost in nursery showed no sign of variation compared to the seed-grown plants. The whole process of culture initiation to plant establishment was accomplished within 5 -6 weeks. A genetic transformation system in J. curcas was established for the first time, using bombardment of particles coated with plasmid pBI426 with a GUS-NPT II fusion protein under the control of a double 35S cauliflower mosaic virus (CaMV) promoter. The β-glucuronidase (GUS) activity in J. curcas shoot apices was significantly affected by the gold particle size, bombardment pressure, target distance, macrocarrier travel distance, number of bombardments, and type and duration of osmotic pre-treatment. The proliferating bombarded shoot apices were screened on medium supplemented with 25 mg dm -3 kanamycin and surviving shoots were rooted on medium devoid of kanamycin. The integration of the transgene into genomic DNA of transgenic plants was confirmed by PCR and Southern blot hybridization. The transgenic plants showed insertion of single to multiple copies of the transgene.

Techniques for the regeneration of Jatropha curcas L. from various explants have been developed. Regeneration from hypocotyl, petiole and leaf explants was evaluated on a range of concentrations of zeatin, kinetin and N 6 benzyladenine (BA) either singly or in combination with indole-3-butyric acid (IBA). Higher regeneration from hypocotyl and petiole explants was obtained on BA with IBA than on zeatin-or kinetin-supplemented media. Leaf discs from the third expanding leaf exhibited higher regeneration potential than those from the fourth leaf. Independent of the explant type, direct adventitious shoot bud induction was recorded highest on MS medium with 2.22 IJ2VI BA and 4.9 ttM IBA. Although the same BA concentration but with reduced IBA concentration (0.49 ttM) proved effective in callus mediated regeneration from hypocotyl and leaf explants, the petioles required lower concentrations of the two growth regulators (0.44 ~VI BA and 0.49 paM IBA). Regenerated shoots could be rooted on growth regulator-free gelled full-strength MS medium. Following simple hardening procedures, the in vitro-raised plants could be transferred to soil and grown to maturity in the field.

Triploid nature of endosperm is the characteristic feature of angiosperms and is formed as a result of triple fusion. Present review discusses the morphogenic response and production of triploid plantlets by endosperm culture. Both mature and immature endosperm used for culture initiation responded differently in cultures. A key factor for the induction of cell divisions in mature endosperm cultures is the initial association of embryo but immature endosperms proliferate independent of embryo. In almost all the parasitic angiosperms, endosperm shows a tendency of direct differentiation of organs without prior callusing, whereas in autotrophic taxa the endosperm usually forms callus tissue followed by differentiation of shoot buds, roots or embryos. The endosperm tissue often shows a high degree of chromosomal variations and polyploidy. Mitotic irregularities, chromosome bridges and laggards are the other important characteristics of endosperm tissues. Triploids are usually seed sterile and is undesirable for plants where seeds are commercially useful. However, in cases where seedlessness is employed to improve the quality of fruits as in banana, apple, citrus, grapes, papaya etc. the induction of triploid plants would be of immense use. Triploid plants have more vigorous vegetative growth than their diploid counterparts. Hence, in plants where the vegetative parts are economically useful, triploids are of good use. This review focuses on the progress achieved so far in endosperm culture to obtain triploid plants.
