Moringa oleifera Protects SH-SY5YCells from DEHP-Induced Endoplasmic Reticulum Stress and Apoptosis

Abstract: Moringa oleifera (MO) is a medicinal plant that has been shown to possess antioxidant, anticarcinogenic and antibiotic activities. In a rat model, MO extract (MOe) has been shown to have a protective effect against brain damage and memory decline. As an extending study, here, we have examined the protective effect of MOe against oxidative stress and apoptosis caused in human neuroblastome (SH-SY5Y) cells by di-(2-ethylhexyl) phthalate (DEHP), a plasticizer known to induce neurotoxicity. Our data show that MOe … Show more

“…24 A previous study showed that after 24 hours of exposure to DEHP, the caspase 3 activity increases in rat pheochromocytoma cells. 25 In our study, caspases 3 & 7 activity was observed (Figure 3) however it was not significantly different compared to the negative control. This low significance can be explained by the variability of the primary cultures that resulted from brain isolated by multiple adult zebrafish and not an established cell line.…”

“…All these results indicated that the MEHP exposure would activate the ER stress. There were some studies have also found MEHP would cause ER stress in other cells like SH‐SY5Y 40 . As an environmental endocrine disruptor, MEHP can cause homeostasis imbalance and increase ROS 41,42 .…”

“…Previous studies suggested that moringa exerts neuroprotective effects against DEHP (di‐[2‐Ethylhexyl] phthalate) a plasticizer linked to neurotoxicity‐induced cytotoxicity in SH‐SY5Y cells. Glucomoringin (GMG) from moringa transformed to isothiocyanate (ITC) on hydrolysis to form GMG‐ITC (Jaafaru et al, 2018) and protects cells by reducing ROS production, blockade of ER stress, and mitochondrial apoptosis (Amara et al, 2021).…”
