A number of monoclonal antibodies have been used to investigate the distributions and rates of lateral motion. of the HLA-A,B, and -DR antigens on several Epstein-Barr virustransformed B-cell lines. The lateral diffusion coefficients (D) of fluorescein conjugates of.the monoclonal antibodies bound to the cell surface were determined by fluorescence recovery after pattern photobleaching. Ds of HLA-A and -B were.found to be comparable and of the order of 1009 to 10-1o cm2/sec for each of the seven monoclonal antibodies and four cell lines examined. The HLA antigens. appear to be monomeric on the cell surface based on experiments using mixtures of arsanilic acid-conjugated and fluorescein-conjugated antibodies. Four monoclonal antibodies against DR antigens were examined. Two ofthese, Genox3.53 and L243, labeled the cell surface uniformly and gave Ds comparable to those obtained for the HILA-A and -B antigens. The other two, DA2 and 2.06, rapidly patched on the cell surface and were immobile. The DA2, L243, and Genox 3.53 antibodies bound outside ofthe caps formed with the arsanilic acid-conjugated 2.06 antibody and a second-step rhodamine-conjugated rabbit anti-arsanilate antibody. The Nethe-lands MHC-class I1 antigens on canine lymphoid cells were detected with monoclonal antibodies (Mabs). On the basis of reactivity with resting or activated T lymphocytes two groups of Mabs were reported earlier. Non-activated T lymphocytes expressed MHC-class I1 antigens recognized by the first group whereas the second group recognized only MHC-class I1 antigens on activated T lymphocytes. In this study we analysed the reactivity pattern of both groups of Mabs with purified canine B lymphocytes. One-and two-colour immunoflowcytometric analysis, immunoprecipitation, immunohistology and MLC inhibition were performed. The Mabs which only reacted with activated T lymphocytes appeared to stain also purified sIg' lymph node cells. Two-colour fluorescence and immunohistology confirmed the reactivity of these Mabs with B lymphocytes. Immunoprecipitation showed the bimolecular structure of these antigens. It was concluded that two subsets of MHCclass I1 antigens can be detected on canine lymphoid cells. One subset showed an aberrant distribution being expressed on both nonactivated and activated T and on B lymphocytes. The second subset of MHC-class I1 antigens could only be detected on B lymphocytes and activated T lymphocytes, a distribution pattern similar to that found in most other species, for instance man.

Two monoclonal antibodies, prepared against a murine B lymphoma and characterized as binding to a cell surface antigen represented primarily on cells ofthe B lineage, were found to bind to human hemopoietic cells. These antibodies recognize similar populations of cells in mice and humans. Antibodies from clones 177.17 and 83.4 bound to 6% of human bone marrow nucleated cells. This included all cells with detectable cell surface Ig (sIg') and those that lack sIg but have detectable cytoplasmic ,u (sIg, cIA+), considered to be the immediate precursors of B lymphocytes (pre-B cells). In addition, these antibodies bound to a subpopulation of T cells and a proportion of null lymphocytes in marrow, spleen, and peripheral blood. An unexpected finding was that established pre-B cell lines were not recognized by these antibodies and possible reasons for this are considered. By using antibody-coated polystyrene plates for cell depletion and recovery, highly enriched preparations of ciA+, sIg-cells have been obtained. These antibodies and enrichment procedures should prove valuable in establishing the minimal requirements for maturation of these putative precursors in vitro, for comparative studies of immunodeficiency/autoimmune diseases in man and experimental animal models, and for monitoring the outcome of therapeutic marrow transplantation.A panel ofmonoclonal rat antibodies was recently prepared and characterized as directed to determinants on murine cells of B cell lineage. One group of seven clones produced antibodies that did not recognize immunoglobulin and that similarly bound to a series of established murine cell lines. Five of these made antibodies ofsatisfactory avidity and were studied in detail. The antigen(s) recognized by these antibodies is displayed on most B cells, a subpopulation of peripheral T cells, and a proportion of antibody-secreting cells (1). In addition, cell surface immunoglobulin negative (sIg-) A number ofcell surface molecules that were originally identified on murine leukocytes have been found to be analogous to human antigens.
