“…It is the first completely validated bioanalytical assay for this chemically very stable drug. The assay is a factor two more sensitive than the only other method using the same analytical technique and protein precipitation [8]. The assay is now successfully implemented for pharmacokinetic-pharmacodynamic relationship evaluation of the drug and the ECG data in the challenge test of patients with a (potential) Brugada syndrome.…”

“…The first assay that was useful for pharmacokinetic evaluation of the drug was reported by Clemans et al [6] using GC-MS after an extremely tedious sample pre-treatment using five liquid-liquid extraction steps and derivatization. Easier assays all use liquid chromatography with fluorescent detection after an ethereal liquid-liquid extraction [7,8], automated solid-phase extraction [9] or on-line extraction [10] procedure. In addition, protein precipitation with acetonitrile was investigated shortly as an alternative for liquid-liquid extraction by Koppel et al [8].…”

“…[1][2][3][4][5][6] But only the verification of sildenafil in tissues fixed in formalin by gas chromatography-mass spectrometry (GC-MS) has been described yet. Consequently, an analysis of ajmaline in histological samples was necessary.…”

“…Different methods for determining ajmaline in serum, blood, urine, and postmortem material are well known. [1][2][3][4][5][6] But only the verification of sildenafil in tissues fixed in formalin by gas chromatography-mass spectrometry (GC-MS) has been described yet. 7 Hence, a method had to be developed to analyze drugs in histological materials by liquid chromatography-tandem mass spectrometry (LC-MS/MS).…”
