Molecular serotyping, virulence gene profiling and pathogenicity of Streptococcus agalactiae isolated from tilapia farms in Thailand by multiplex PCR

Abstract: We identified two major serotypes of S. agalactiae isolates associated with the outbreak in tilapia culture in Thailand. We developed multiplex PCR assays for 14 virulence genes, which may be used to predict the pathogenicity of the isolates and track future infections. Multiplex PCR typing of the GBS virulence genes was developed and might be further used to predict the pathogenicity of S. agalactiae.

“…The experimental fish were anesthetized with clove oil to minimize stress during vaccination and challenge testing. S. agalactiae serotypes Ia and III were cultured as described previously 7 . S. agalactiae serotype III was used for polyclonal antibody (pAb) production, which was kindly provided by Prof. Ikuo Hirono, TUMSAT, Japan.…”

“…Serotype Ib strains isolated in Central and South America are ST-260 and ST-552 in CC552 (27, 29) and strains isolated in Australia, Israel, Belgium, China, Ghana, USA and Southeast Asia belong to ST-261 (5, 6, 9, 13, 2931). Serotype III is commonly causative of disease in humans, but has also been isolated from fish in Thailand, China and recently in Brazil (26, 3234).…”

“…GBS was initially described in 1887 as an animal pathogen responsible for mastitis in ruminants and although largely eradicated in some countries with a highly developed dairy industry, remains a major cause of bovine mastitis in many countries 1 . In 1966, GBS infection of fish was reported in the USA 2 with subsequent outbreaks described in fish farms in Asia and the Americas 3,4 . GBS infection is often associated with high levels of mortality in fish and with the worldwide growth of aquaculture has significant economic impact 5 .…”
