“…DISCUSSION We recently reported the occurrence of a membrane-bound regulatory protein(s) that mediates the stimulation of adenylate cyclase by Gpp(NH)p and NaF (1). This protein(s) is not solubilized with 0.1 mM EDTA, 0.5-1 M KCI, or 4 M urea (unpublished data but, after detergent solubilization, it can reconstitute the Gpp(NH)p and NaF sensitivity of adenyl~te cyclase preparations that have been previously rendered nonresponsive to these activators (1). The action of this regulatory protein(s) does not exhibit tissue or species specificity.…”

“…We have reported (1) the development of such a technique which involves the preparation of a Gpp(NH)p-and NaF-insensitive brain particulate adenylate cyclase by differential detergent extraction. The enzyme regained its characteristic Gpp(NH)p and NaF responses upon the addition of one or more proteins solubilized from membrane preparations of various tissues (1). These regulatory proteins can be partially separated from solubilized adenylate cyclase by gel filtration on Ultrogel AcA 34 columns.…”

“…The gradients contained 25 mM Tris-HCl (pH 7.6), 0.1% Triton X-100 (all detergent concentrations are expressed as wt/vol), and 5 mM MgCl2. Samples (0.2 ml) were loaded and the gradients were centrifuged in a Beckman SW 50.1 rotor at 45,000 rpm for [14][15][16] hr at 40C, and 0.2-ml fractions were collected. Gel permeation chromatography was performed by applying 0.5-ml samples to 26-ml Ultrogel AcA-34 columns and elutingwith 50 mM Tris-HCl, pH 7.6/0.1% Triton X-100/5 mM MgCl2; 0.8-ml fractions were collected.…”

“…The response of adenylate cyclase from mammalian brain to 5'-guanylyl imidodiphosphate (p[NH]ppG) can be abolished by two different procedures (7,8) and subsequently restored by addition of a diluted unfractionated detergent extract of brain that contains both the CU and the G/F. Because the G/F is not separated from the CU, the interpretation of the experiments is complicated.…”
