Molecular recognition of DNA by small molecules and proteins is a fundamental problem in structural biology and drug design. Understanding of recognition in both sequence-selective and sequence neutral ways at the level of successful prediction of binding modes and site selectivity will be instrumental for improvements in the design and synthesis of new molecules as potent and selective gene-regulatory drugs. Minor groove is the target of a large number of non-covalent binding agents. DNA binding with specific sequences, mostly AT, takes place by means of a combination of directed hydrogen bonding to base pair edges, van der Waals interactions with the minor groove walls and generalized electrostatic interactions. These factors are also responsible for protein-DNA recognition, and a number of unifying rules governing the interactions have been elucidated although it has been realized that the earlier goal of a simple recognition code between amino acids and bases is not attainable. At present relatively little is understood about the mode of action at the molecular level of the majority of minor groove-interacting drugs, although there is increasing evidence that they may act by directly blocking or inhibiting protein-DNA recognition. The present review has the aim to focus on interactions between minor groove binders and DNA through a variety of techniques that are commonly used to analyze the DNA binding properties of small molecules. In fact in the last years several articles dealing with in silico techniques on DNA minor groove binders (molecular modeling, molecular dynamics, QSAR) have been published. All these studies can be considered a support in defining valid predictive models. For this reason a compendium of all matter could be an useful support for future developments.

is commemorating its 25 th anniversary, and this paper is intended to draw an overview of the Brazilian Medicinal Chemistry over all these years. In 1977 Brazil had almost no activities at all in the field, albeit many efforts were already on the way for encouraging Brazilian Scientists to enter the area. Among many different endeavours to help medicinal chemists to fulfil their proposals and the establishment of an ongoing research with the help of networks, the Sociedade Brasileira de Química created, in 1991, its own Division on Structure and Activity Relationship, which became the Division of Medicinal Chemistry, in 1997.

This paper describes 2D-QSAR and 3D-QSAR studies against Candida albicans and Cryptococcus neofarmans for a set of 20 bisbenzamidines. In the studies of 2D-QSAR with C. albicans it was obtained a correlation between log MIC -1 and lipolo component-Z (r 2 = 0.68; Q 2 = 0.51). In the case of C. neofarmans a correlation between log MIC -1 and lipolo component-Z and of Balaban index (r 2 = 0.85; Q 2 = 0.6) was obtained. 3D-QSAR studies using CoMFA showed that the steric fields contributed more to the predicted activities for Candida albicans (94.9%) and Cryptococcus neofarmans (97.9%).Keywords: QSAR; bis-benzamidines; antifungal activity. INTRODUÇÃOAs infecções causadas por fungos são, geralmente, limitadas à superfície dos tecidos e são relativamente fáceis de serem curadas. 1,2Com os avanços da medicina, o número de pacientes em internação hospitalar prolongada cresceu muito, assim como a população de pacientes imunologicamente vulneráveis a fungos e bactérias, levando a um aumento da frequência de infecções causadas por patógenos oportunistas. Tais internações propiciam uma maior exposição do paciente a patógenos multirresistentes, comumente presente nos centros hospitalares.3,4 Adicionalmente, em pacientes imunossuprimidos as infecções causadas por fungos podem ser graves. 5,6 Ao longo dos últimos 10 anos, a incidência de infecções causadas por fungos tem aumentado muito em número e em gravidade. Para piorar este grave quadro epidemiológico, o uso incorreto de antibióticos de amplo espectro de ação tem levado ao surgimento de populações de microorganismos resistentes a estes agentes quimioterápicos, 7 apesar da maior parte dos fungos patogênicos serem muito bem conhecidos tanto quanto o seu tratamento clínico.3-9 O tratamento destas infecções fúngicas invasivas (IFIs) e oportunistas permanece ainda insatisfatório com o uso da maior parte dos agentes antifúngicos usuais, tais como itraconazol e anfotericina B, e além disso os agentes patogênicos oportunistas têm aumentado cada vez mais a sua resistência mesmo com o uso de novos agentes antifúngicos como, por exemplo, o voriconazol.
