Molecular physiology reveals ammonium uptake and related gene expression in the seagrass Zostera muelleri

Abstract: Seagrasses are important marine foundation species, which are presently threatened by coastal development and global change worldwide. The molecular mechanisms that drive seagrass responses to anthropogenic stresses, including elevated levels of nutrients such as ammonium, remains poorly understood. Despite the evidence that seagrasses can assimilate ammonium by using glutamine synthetase (GS)/glutamate synthase (glutamine-oxoglutarate amidotransferase or GOGAT) cycle, the regulation of this fundamental metabo… Show more

“…Zostera muelleri GOIs were newly designed using Z. muelleri database from AquaticPlantsDB R (Sablok et al, 2018), 2 while housekeeping genes (HKGs) were taken from previous studies (Schliep et al, 2015;Pernice et al, 2016;Kim et al, 2018). For P. australis, however, no molecular resources are available to date, thus selected GOIs and HKGs were either newly designed or taken from previous studies on the congeneric species P. oceanica.…”

“…stress related genes as part of plant stress response (Kan et al, 2017). It has been suggested that regulation of genes in this pathway could be used as an indicator for exposure to ammonium pulses in seagrass (Pernice et al, 2016). Passive ammonium transport is not regulated (Touchette & Burkholder, 2000), therefore uptake of ammonium by the plant continues.…”

“…While the catalase gene has been reduced to a single copy in Z. marina (possibly due to reduced xylemcharacteristics of submerged plants), whilst all three types of superoxide dismutase remain (Olsen et al, 2016 ). Glutathione-related transcripts have been isolated and profiled in P. oceanica, Z. muelleri , and Z. marina (Massa et al, 2011 ; Lauritano et al, 2015 ; Olsen et al, 2016 ; Pernice et al, 2016 ). In protein, enzymes related to cellular stress and photosynthesis genes in which the studies of seagrass in response to differing light conditions also revealed the differential expression of genes for (Dattolo et al, 2013 , 2014 ).…”
