University of Manitoba, WinnipegThymocytes collected from spleens of lethally irradiated and thymocytereconstituted mice, 6 h after "education" with SRBC, exert a markedly augmented helper cell function, especially for the 7 S response. Pretreatment of primed thymocytes with a rabbit anti-mouse Ig serum and complement abolished the augmented helper cell function, although it had no effect on unprimed cells. Treatment of 6 h primed T cells with an anti-0 serum and complement also abolished the augmented helper cell function. However, in striking contrast with unprimed T cells where such treatment entirely abolished the helper cell function, the anti-0 treated 6 h T cells are still able to provide a significant helper cell function of the same magnitude as untreated nonprimed cells. This result suggests that a T cell subpopulation 6 h after priming becomes resistant t o lysis by anti-0 serum and supports a substantial helper cell function. Since the sum of plaque-forming cells (PFC) given by anti-Ig and anti-@treated 6 h primed cells was far smaller than the PFC given by untreated cells, a T-T synergism is implicated as a mechanism for the augmented helper cell function of 6 h primed thymocytes.The amplification was also eliminated following treatment with a mouse anti-SRBC serum indicating that some primed cells carry antigen on their surface. The data from various recombinations of primed cells treated with one of these three antisera, as well as primed cells treated sequentially with various combinations of two of the three antisera, suggested that the Ig' and antigen-carrying cells belong t o the same subpopulation which is resistant t o lysis by anti-0 serum. These cells are distinct from the rest which remain Ig-and still sensitive t o lysis by anti-Q serum (0').The Ig and antigen most likely represent a cytophilic complex present on the surface of a subpopulation of T cells. Thymocytes collected 5 days after education show little or no synergistic effect.The T-T cell synergism described here may represent a basic regulatory function of T cells.

A large dose of tumour cells (10(7)) from two different tumours produced a significant increase of Ig-bearing spleen cells 6 h after intraperitoneal administration in syngeneic mice, like conventional antigens previously tested. Incubation of normal spleen cells in serum taken 6 h after administration of tumour cells reproduced in vitro the changes observed in the spleen cell population of the serum donors, demonstrating the presence of a cytophilic Ig, which was shown to be taken up by T cells. Serum collected 6 h after tumour cell inoculation contains also a 4S factor which can generate in vitro cytophilic Ig for T cells in the presence of a foreign soluble protein or tumour antigen. Extracts obtained from tumours cells, which were shown to contain tumour antigens, were labelled with 125I and mixed with the 4S factor and normal 7S Ig. Upon fractionation of the mixture, high molecular weight material containing radioactivity and 7S Ig were eluted in the void volume, well ahead of the position of both the tumour cell preparation and the 7S Ig. This material contained Ig cytophilic for splenic T cells and it is likely that it represents cytophilic complexes of Ig and tumour antigens. It is postulated that the cytophilic Ig detected in the serum of animals 6 h after injection of a large number of tumour cells represents cytophilic complexes of Ig and tumour antigens formed through the mediation of a soluble factor.

Inhibition of antigen absorption by the immune gut 701 Interference of oral immunization with the intestinal absorption of heterologous albuminRats were immunized with human serum albumin (HSA) by a single intragastric administration o f 200 mg of HSA. Two weeks later their capacity to absorb a similar intragastric test dose of HSA was found to be greatly impaired, the concentrations of HSA in mesenteric venous serum having been reduced to 33 %, 25 % and 60 5% of those in similarly tested but unprimed animals, respectively 1, 2 and 3 hours after the intragastric test dose. Rats given antigen (HSA) together with intestinal secretions from intragastricimmunized rats also showed a striking decrease in intestinal absorption of a test dose, compared t o controls given the antigen together with nonimmune intestinal secretions.From these data it is concluded that local immunization of the gut impairs its capacity to absorb the corresponding antigen, and that this effect is largely due t o the failure t o absorb antigen bound t o secretory antibodies.
