Molecular Epidemiology of Human Astrovirus Diarrhea among Children from a Periurban Community of Mexico City

Abstract: Human astroviruses (HAstVs) were detected in 23 stool samples from 365 diarrhea episodes among 214 children (<18 months old) prospectively monitored for diarrhea in Mexico City. Stool samples were tested by EIA and reverse transcriptase-polymerase chain reaction (RT-PCR) analysis. EIA was less sensitive (74%) and equally specific, compared with RT-PCR analysis using type-common primers for HAstV detection. Of 31 HAstV isolates, EIA typed 18 (69%) of 26 EIA-positive samples, and RT-PCR analysis typed 26 (84%) o… Show more

“…The infection rate ranged from 2 to 6 % for hospitalized children with acute diarrhoea and 5-17 % for acute diarrhoea cases from the community [3]. In both developed and developing countries, HAstV infection was associated with diarrhoea episodes and ranged from 3 to 12 and 2 to 26 % respectively [18,23,29,34,40,41].…”

“…Recombination events have been observed in astroviruses which infect both human and animals. Novel recombinant human astrovirus strains were first described in children with HAstV_G3 (ORF1b) and HAstV_G5 (ORF2) combination in the ORF1b/ORF2 junction [40]. Human astrovirus wild-type strain obtained from a Kenyan child showed genetic recombination event in the sequence analyses of ORF1a (HAstV_G6/7), ORF1b (HAstV_G3) and ORF2 region (HAstV_G2) [42].…”

“…These small, spherical (28-30 nm in diameter), non-enveloped viruses have a plus sense, single-stranded polyadenylated RNA (ssRNA) 13 genome of about 7 kb in length . To date, eight HAstVs genotypes (HAstV-1 to HAstV-8) have been described, and HAstV-1 has been reported as the most prevalent type in 6,46 both developing and developed countries .…”

“…The HAstV genome is a polyadenylated plus-strand RNA molecule of ϳ7 kb organized in three open reading frames (ORFs): ORF1a and ORF1b, at the 5Ј end of the genome, code for the nonstructural viral proteins, while ORF2, at the 3Ј end, encodes the capsid proteins (8). Based on the nucleotide and encoded amino acid sequences of either the amino-terminal or carboxy-terminal region of ORF2, these viruses have been grouped into eight genotypes (11,16), which have been shown to completely correlate with the eight established HAstV serotypes (9,10,12,24,28; J. B. Kurtz and T. W. Lee, Letter, Lancet ii:1405, 1984).…”
