Molecular Detection of Spotted Fever Group Rickettsia in Feral Raccoons (<i>Procyon lotor</i>) in the Western Part of Japan

Abstract: ABSTRACT. Rickettsial infection in feral raccoons (Procyon lotor) in the western part of Japan (Shimane, Fukuoka, Saga and Nagasaki Prefectures) was surveyed by a nested polymerase chain reaction (PCR) assay detecting the rickettsial citrate synthase (gltA) gene. Four of one hundred and ninety-four feral raccoon spleens (2.1%) were positive for Rickettsia spp. One gltA gene sequence was identical to R. helvetica, whereas the other 3 sequences were identical and had the highest similarity (98.4%) to R. amblyomm… Show more

“…Proteus is mainly used as diagnostic test for Rickettsia because a cross-reaction exists. Rickettsia has been reported in raccoons from the US and Japan (Alexander et al 1972, Baba et al 2013 and in humans from the northern and southeastern regions of Mexico (Zavala-Castro et al 2006). Rickettsial infections were associated with tick bites, but in the present study antibodies against Proteus were not detected.…”

“…asiatica and Candidatus R. tarasevichiae, have also been reported 13,14 . In addition, several studies conducted in Japan have documented the presence of other Rickettsia species/genotypes in animals and questing ticks 1517 . However, in most cases, only single or a limited number of genes have been analysed, making it difficult to generate an overview of the genetic diversity of SFG rickettsiae, since multiple gene sequencing are recommended in the classification of rickettsial isolates 18 .…”

“…In two studies in Japan, molecular detection of Rickettsia spp. in raccoons was performed [ 80 , 81 ]. Rickettsia ( R. ) helvetica and R felis were among the identified species, with prevalences between 0.1 and 1.6% [ 80 , 81 ].…”
