Molecular comparison of topotypic specimens confirms Anopheles (Nyssorhynchus) dunhami Causey (Diptera: Culicidae) in the Colombian Amazon

Abstract: The presence of Anopheles (Nyssorhynchus) dunhami Causey in Colombia (Department of Amazonas) is confirmed for the first time through direct comparison of mtDNA cytochrome c oxidase I (COI) barcodes and nuclear rDNA second internal transcribed spacer (ITS2) sequences with topotypic specimens of An. dunhami from Tefé, Brazil. An. dunhami was identified through retrospective correlation of DNA sequences following misidentification as Anopheles nuneztovari s.l. using available morphological keys for Colombian mos… Show more

“…lesteri from Korea and An. paraliae from Thailand, based on ITS2 sequences, was in accordance with previous reports of different groups of Anopheles, e.g., the Anopheles gambiae complex (0.4-1.6%) (Paskewitz et al 1993), Anopheles dunhami and Anopheles nuneztovari (mean genetic distance = 0.025) (Ruiz et al 2010), Anopheles fluviatilis S and An. minimus C (pairwise distance = 0.036) (Singh et al 2006), Anopheles kunmingensis and Anopheles liangshanensis (pairwise distance = 0.0381) and An.…”

“…dunhami in Amazonian Brazil, and its range may be more extensive than previously believed. The low genetic distance found between these two species (mean = 2.6%) and results of previous studies, including multiple markers, 37 isozymes, 38 ITS2, COI and white gene sequences, 18,39 strongly suggest recent divergence. Given that An.…”

“…The rDNA ITS2 region was amplified using the primers of Collins and Paskewitz (1996) and the polymerase chain reaction (PCR) conditions described by Linton et al (2001). A portion (710 bp) of the barcoding region of COI (mDNA) was amplified using the primers designed by Folmer et al (1994) and the PCR conditions described by Ruiz et al (2010). The products were visualised on 1% agarose gel containing 0.5 mg/ mL of ethidium bromide.…”
