Molecular cloning, structural analysis, and expression of carp ZP3 gene

Abstract: Two types of cDNAs coding for a major component of carp egg membrane were clones from a carp ovarian cDNA library. They encode polypeptides of 422-424 amino acid residues whose sequences are homologous to those of medaka and mammalian ZP3. Similar to the mammalian ZP3 genes, carp ZP3 gene also consists of eight exons and seven introns. Carp ZP3 genes are 2.9 kb in length and present in multiple forms. Carp ZP3 is a glycoprotein of 45 kDa. It was transcribed and translated exclusively in oocytes, in contrast wi… Show more

“…For example, rainbow trout, cod, and Atlantic salmon [18,19,48], medaka, Oryzias latipes [49-51], winter flounder, Pseudopleuronectes americanus [52], and gilthead seabream, Sparus aurata [53], synthesize Zr-protein in the liver. Other species, such as carp, Cyprinus carpio [54,55], and pipefish, Syngnathus scovelli [56] appear to synthesize Zr-protein in the ovary. Hence, the primary sequence of Zr-proteins is known in many teleost species, including winter flounder [52], medaka [49,50], carp [54,55], Atlantic salmon [48], and rainbow trout [57-59].…”

“…All euteleost fish and Clupeiformes, which is diverged early from Otocephala, have both ovarian‐ and liver‐expressing ZP genes (Sano et al., ). The liver‐expressing ZP genes in Otocephala have lost the evolutionary process to Ostariophysi, and the egg envelopes of Ostariophysi, such as milkfish, zebrafish, carp, and goldfish, are composed of ovarian‐synthesized ZP proteins (Chang et al., , ; Sano et al., , ; Wang et al., ). The egg envelopes of euteleost species have been particularly well studied, and their egg envelopes are principally composed of liver‐synthesized ZP proteins (Hamazaki et al., , Murata et al., ).…”

“…The molecular composition of invertebrate egg ECMs appears to differ significantly from that of vertebrate egg ECMs. In addition, ZP protein homologues have been identified in several nonmammalian species, including the frog [27][28][29], chicken [30,31], and even fish [32][33][34][35][36], which diverged from mammals 650 million years ago. Instead, invertebrate VEs are composed of completely different proteins, such as VERL in the abalone [22] and the 350-kDa receptor for sperm, VE-B, and other proteins in the sea urchin [23][24][25].…”
