Molecular cloning, polymorphism, and functional activity of the bovine and water buffalo Mx2 gene promoter region

Abstract: BackgroundBovine Mx2 gene sequences were already reported, but further information about the gene properties is not yet available. The objective of the current study was to elucidate the structural properties of the bovine Mx2 gene mainly the promoter region and its possible functional role. If available, such information would help in assessing the functional properties of the gene, which was reported to confer antiviral action against recombinant VSV.ResultsExaminations on the bovine genomic BAC clone—confir… Show more

“…BAMBI gene was cloned into PMD-18-T vector (Takara Biotechnology Co., Ltd., Dalian, China) and sequenced to identify its sequence according to previous report ( 28 ). BAMBI gene was subsequently cloned into eukaryotic expression vector pCMVp-NEO-BAN (pBAMBI; Takara Biotechnology Co., Ltd.) to generate BAMBI-overexpressed BGC-823 cells.…”

“…During early pregnancy in cattle, conceptus IFNT induces MX2 expression in circulating PBL and may be used to diagnose pregnancy [64,66]. The bovine MX2 promoter region contains multiple ISREs which would explain why MX2 is highly expressed in bovine tissues in response to type I IFNs [67].…”

“…However, considering that the IFN promoter sequences in human and trout are different [27,[41][42][43], and since EPC cells were grown at 15°C, while HeLa and FHs74 cells were grown at 37°C, one possibility could be that temperature-dependent conformation of the Mx promoter region interfered with activation of transcription in human cell lines... Although there are currently no concerns about consumers eating DNA-vaccinated fish each vaccine construct will be evaluated on an individual basis [44] and the lack of activity of the trout Mx promoter in human cells could make it attractive for some fish DNA vaccines.…”
