Molecular Cloning of Peroxinectin Gene and Its Expression in Response to Peptidoglycan andVibrio Harveyiin Indian White ShrimpFenneropenaeus Indicus

Abstract: The cDNA sequence of peroxinectin was obtained from the haemocytes of Indian white shrimp Fenneropenaeus indicus using RT-PCR and RACE. Fenneropenaeus indicus peroxinectin (Fi-Pxn) sequence has an open reading frame (ORF) of 2415 bp encoding a protein of 804 amino acids with 21 residues signal sequence. The mature protein has molecular mass of 89.8 kDa with an estimated pI of 8.6. Two putative integrin-binding motifs, RGD and KGD, were observed at the basic N-terminal and C-terminal part of the mature aminoaci… Show more

“…Among the immunity-related genes, proPO is an important constituent of the proPO cascade reaction that plays an important role in invertebrate protection against invading pathogens by melanization of pathogens (70, 71). Peroxinectin (PXN) is another defense-related gene and is a cell adhesive protein known to be strongly associated with the proPO system (72). It regulates the expression of antimicrobial peptides (AMPs) in invertebrates (73, 74).…”

“…The resulting hemocyte pellet was used for total RNA isolation by following the method of TRI‐Reagent (Invitrogen). The full length sequence of Fein ‐PX gene was determined by degenerated primer designing, reverse transcriptase‐PCR, cloning and sequencing of overlapping PCR, and rapid amplification of cDNA ends method (Shanthi et al , ). The Fein ‐PX gene sequence was submitted to NCBI GenBank with accession no.…”

“…The presence of integrin binding motif (LDV) in AsHPX2 protein suggested its peroxinectin-like nature. Peroxinectins are invertebrate peroxidases that are involved in cell adhesion (Liu et al, 2004;Vizzini et al, 2013;Shanthi et al, 2014). Interestingly, the surface orientation of LDV motif in AsHPX2 protein seems to be facilitating its adhesion in a way similar to other adhesion molecules (Johanssen et al, 1999;Liu et al, 2005).…”
