Molecular cloning of a human Ca2+-dependent cell-cell adhesion molecule homologous to mouse placental cadherin: its low expression in human placental tissues.

Abstract: Abstract. P-cadherin is a subclass of Ca2+-dependent cell-cell adhesion molecules present in mouse placenta, where its localization suggests a function of connecting the embryo to the uterus (Nose, A., and M. Takeichi. 1986. J. Cell Biol. 103:2649-2658. We recently identified a human cadherin detected by an mAb capable of disrupting cell-cell adhesion of A-431 cells, and found that it was closely related immunochemically to mouse P-cadherin. Curiously, this cadherin was undetectable in human placenta by immuno… Show more

“…Human ␤-actin cDNA was radiolabeled using Random Primed DNA Labeling kit (Boehringer Mannheim). Human multiple tissue Northern blot I and human brain multiple tissue Northern blots II and III (Clontech) were hybridized with radiolabeled probes (22). Filters were washed in 2 ϫ SSC (1 ϫ, 150 mM NaCl and 15 mM sodium citrate), 0.05% sodium dodecyl sulfate for cadherin-14 cDNA and in 0.1 ϫ SSC, 0.1% sodium dodecyl sulfate for ␤-actin cDNA at 65°C for 60 min as described previously (22).…”

“…cDNAs for human N-cadherin, human P-cadherin, mouse R-cadherin and human cadherin-11 were kind gifts from Dr. Avri Ben Ze'ev [24], Dr. Setsuo Hirohashi [25], Dr. Masatoshi Takeichi [26] and Dr. Akira Kudo [27], respectively. Mutant E-cadherin (Ep120AAA) that cannot bind to p120 has been described [8].…”

“…RNA blottings were performed as described previously (35). Premade filters (human brain multiple tissue Northern blots II and III and human muscle multiple tissue Northern blot) purchased from CLONTECH were also used.…”

“…E-, N-, and P-cadherins and cadherin-4, -5, -6, -8, -11 (OB-cadherin), and -12 (3,29,30,31,(35)(36)(37). We are interested in how many classic cadherin molecules actually exist in humans and how these molecules function and cooperate in the development and maintenance of the integrity of the human body, as well as in various pathogenic states such as cancers.…”
