Molecular cloning and functional characterization of  the genes encoding benzoate and p-hydroxybenzoate degradation by the halophilic Chromohalobacter sp. strain HS-2

Abstract: Chromohalobacter sp. strain HS-2 was isolated from salted fermented clams and analyzed for the ability to grow on benzoate and p-hydroxybenzoate as the sole carbon and energy source. HS-2 was characterized as moderately halophilic, with an optimal NaCl concentration of 10%. The genes encoding the benzoate metabolism were cloned into a cosmid vector, sequenced, and then analyzed to reveal the benzoate (benABCD) and catechol (catBCA) catabolic genes, both of which are flanked on either side by LysR-type transcri… Show more

“…This study showed that the organism degraded benzoate and salicylate to catechol and then to cis, cis-muconate thus indicating degradation via the ortho-cleavage pathway. Kim et al [130] have isolated a Chromohalobacter sp. strain HS-2 from salted fermented clams that degrades benzoate and p-hydroxybenzoate as the sole carbon and energy sources.…”

“…After growth on hydroxybenzoic acids, additional cleaving dioxygenase is usually induced (26). 4-Hydroxybenzoic acid as well as vanillic acid were found to induce mainly protocatechuate 3,4-dioxygenase (7,13,14,16,21). Gentisate dioxygenase was also found in the cells grown on hydroxybenzoic acids (6,27).…”

“…Halomonas campisalis grown on benzoate or salicylate revealed activity on 1,2-CAT (Oie et al 2007). Kim et al (2008) showed the presence of 1,2-CAT and 3,4-PCA in a benzoate-and phydroxy-benzoate-metabolizing halophile, Chromohalobacter sp. strain HS-2.…”
