S-Adenosylmethionine decarboxylase (SAMDC) is a key rate-limiting enzyme involved in polyamines biosynthesis, and it plays important roles in plant growth, development and stresses response. However, no SAMDC gene was reported in rubber tree. Here we report characteristics of an SAMDC gene (HbSAMDC1) in rubber tree. HbSAMDC1 contains a 1080 bp open reading frame (ORF) encoding 359 amino acids. Quantitative real-time PCR analyses revealed that HbSAMDC1 exhibited distinct expression patterns in different tissues and was regulated by various stresses, including drought, cold, salt, wounding, and H 2 O 2 treatments. HbSAMDC1 5 0 untranslated region (UTR) contains a highly conserved overlapping tiny and small upstream ORFs (uORFs), encoding 2 and 52 amino acid residues, respectively. No introns were located in the main ORF of HbSAMDC1, whereas two introns were found in the 5 0 UTR. In transgenic tobaccos, the highly conserved small uORF of HbSAMDC1 is found to be responsible for translational repression of downstream bglucuronidase reporter. To our knowledge, this is the first report on molecular cloning, expression profiles, and 5 0 UTR characteristics of HbSAMDC1. These results lay solid foundation for further elucidating HbSAMDC1 function in rubber tree.

The biosynthesis of the polyamines spermidine (Spd) and spermine (Spm) from putrescine (Put) is catalysed by the consequent action of two aminopropyltransferases, spermidine synthase (SPDS EC: 2.5.1.16) and spermine synthase (SPMS EC: 2.5.1.22). Two cDNA clones coding for SPDS and SPMS homologues in the nitrogen-fixing nodules of the model legume Lotus japonicus were identified. Functionality of the encoded polypeptides was confirmed by their ability to complement spermidine and spermine deficiencies in yeast. The temporal and spatial expression pattern of the respective genes was correlated with the accumulation of total polyamines in symbiotic and non-symbiotic organs. Expression of both genes was maximal at early stages of nodule development, while at later stages the levels of both transcripts declined. Both genes were expressed in nodule inner cortical cells, vascular bundles, and central tissue. In contrast to gene expression, increasing amounts of Put, Spd, and Spm were found to accumulate during nodule development and after maturity. Interestingly, nodulated plants exhibited systemic changes in both LjSPDS and LjSPMS transcript levels and polyamine content in roots, stem and leaves, in comparison to uninoculated plants. These results give new insights into the neglected role of polyamines during nodule development and symbiotic nitrogen fixation (SNF).

The expression of MdSAMDC2 gene, which encodes S-adenosylmethionine decarboxylase (SAMDC) in apple, was up-regulated by low temperature, salt and drought stresses. To identify its in vivo biological functions in the responses to stresses, the promoter region of MdSAMDC2 was isolated and characterized by analyzing the cis-acting regulatory elements and GUS reporter gene by an Agrobacterium-mediated transformation. GUS activity was enhanced upon salt and cold stresses, indicating that MdSAMDC2 promoter region controls gene transcription under stresses. In parallel, several lines of the transgenic tobacco plants over-expressing MdSAMDC2 were obtained. The contents of three polyamines greatly increased in the transgenic lines compared with the non-transgenic WT control. Upon exposed to low temperature (4 °C), salt (150 and 250 mM NaCl) and osmotic (20 % polyethylene glycol) stresses, transgenic plants produced more free polyamines and more active antioxidative enzymes such as superoxide dismutase and catalase than the WT control. Meanwhile, malondialdehyde content, an indicator for membrane lipid peroxidation, decreased in transgenic plants relative to the WT control. Thus, over-expression of MdSAMDC2 in tobacco conferred tolerance to stresses.
