Molecular Characterization of Vitex agnus-castus L. (Verbenaceae) Populations Grown in Aydin, Turkey

Abstract: In this study, we performed a genetic diversity analysis using RAPD markers for some Vitex agnus-castus populations grown in Aydin, Turkey. Total genomic DNA isolation from the leaves of Vitex agnus-castus was performed using a commercial kit. Seven RAPD primers (OPA-02, OPA-05, OPA-13, OPA-15, OPA-16, OPA-18, OPA-20) were used to determine genetic diversity among populations. Polymerase Chain Reaction (PCR) was performed with all genomic DNA samples and primers. PCR products were run in agarose gel electropho… Show more

“…6). In the study of Sevindik et al (2019), Çakmar purple flowered and İncirliova purple flowered populations were found in a separate group, while Koçarlı white flowered, Germencik purple flowered, Çine purple flowered and Çakmar white flowered populations were found in the same group. Subclade B consisted of Çakmar pink flowered, Çine pink flowered, Erbeyli white flowered and Köşk purple flowered, and this group was supported with a 60 % bootstrap value (Fig.…”

“…The closest genetic distances were between Çine pink flower and Çakmar purple flower, and Çakmar pink flower and Çakmar purple flower populations with a value of 0.05556. RAPD markers are useful tools for determining genetic relationships among Vitex agnus-castus genotypes (SEVINDIK et al, 2019). Phylogenetic analysis were performed using seven RAPD primers in 13 populations of 6 species belonging to Conringia spp.…”

“…Besides inexpensive and easy-to-use properties, RAPD-PCR (random amplified polymorphic DNA-polymerase chain reaction) is used in determination of random genomic regions linked to certain plant characteristics (WELSH and MCCLELLAND, 1990;WILLIAMS et al, 1990). RAPD analysis was used with molecular characterization in Vitex agnuscastus L.(Verbenaceae) (SEVINDIK et al, 2019), genetic diversity in Sorghum bicolor (RUIZ- CHUTÁN et al, 2019), and genetic variation and molecular relationships in Conringia Heist. ex Fabr.…”
