Fusarium strains were isolated from rotten banana fruit imported into Hungary from some African and some Neotropical countries. The strains were identified using morphological features, 2-benzoxazolinone tolerance, translation elongation factor (EF-1α) sequences and inter simple sequence repeat (ISSR) analysis. All strains from Africa proved to be F. verticillioides whereas the strains from the Neotropics are Fusarium musae. According to the PCR proof and the fumonisin toxin measurement F. musae strains cannot produce any fumonisins (FB1–4).

ABSTRACT. We optimized RAPD techniques by increasing the length of RAPD primers and performing a strict screening of PCR annealing temperature to distinguish 60 sweet orange cultivars from the Research Institute of Pomology at the Chinese Academy of Agricultural Sciences. A new approach called cultivar identification diagram (CID) was used to improve the efficiency of RAPD markers for cultivar identification. Thirteen effective primers were first screened from 54 RAPD arbitrary 11-mer primers based on their amplification products and amplified polymorphic bands; they were then used for PCR amplification of all 60 cultivars. All cultivars were manually and completely separated by the polymorphic bands appearing in DNA fingerprints from 13 primers; a CID of the 60 sweet orange cultivars was then constructed. This CID separated all the cultivars from each other, based on the polymorphic bands; the corresponding primers were marked in the correct positions on the sweet orange CID. The CID strategy facilitates the identification of fruit cultivars with DNA markers. This CID of sweet orange cultivars will be very useful for the protection of cultivar rights and for early identification of seedlings in the nursery industry.

Fusarium oxysporum f. sp. pisi (FOP) is a significant and destructive pathogen of field pea in Algeria. In the present study, 50 isolates of F. oxysporum f. sp. pisi, the causal agent of pea (Pisum sativum) wilt, collected from different parts of western Algeria and representing four races of the pathogen, were analyzed for virulence. The wilt incidence ranged from 6.66 to 88.33% on a highly susceptible cultivar (Little Marvel). Twenty-one isolates belonging to four races of FOP and one nonpathogenic F. oxysporum (FO) isolate were analyzed for vegetative compatibility in order to reveal the genetic structure of the population and to check the reliability of the method for the identification of physiological races of FOP. Obtained results showed that the FOP isolates could be classified into four main vegetative compatibility groups (VCGs) that corresponded to races l, 2A, 2B and 5. The race 6 isolate fell into the race 1 VCG. To our knowledge, this is the first such study in Algeria of its kind.
