Molecular Characteristics of Cell Pyroptosis and Its Inhibitors: A Review of Activation, Regulation, and Inhibitors

Abstract: Pyroptosis is an active and ordered form of programmed cell death. The signaling pathways of pyroptosis are mainly divided into canonical pathways mediated by caspase-1 and noncanonical pathways mediated by caspase-11. Cell pyroptosis is characterized by the activation of inflammatory caspases (mainly caspase-1, 4, 5, 11) and cleavage of various members of the Gasdermin family to form membrane perforation components, leading to cell membrane rupture, inflammatory mediators release, and cell death. Moderate pyr… Show more

“…GSDMD is a highly sought-after drug target, although the development of specific GSDMD inhibitors was not successful to date [44][45][46][47] . Therapeutic application of the potent pyroptosisinhibiting nanobodies described in this study would require efficient delivery of nanobodies into the cytosol of target cells.…”

“…It has two signaling pathways: a canonical pathway mediated by cysteine-1 and a non-canonical pathway mediated by cysteine-11 (Rühl et al, 2018). It is characterized by the activation of inflammatory cysteine-1 enzymes and cleavage of various members of the gasdermin family to form membrane-perforating, leading to cell membrane rupture, the release of inflammatory mediators and cell death ultimately (Wei et al, 2022). Autophagy refers to the catabolic process by which a cell turns over its own components (Yang and Klionsky, 2010).…”

“…Physiologically, pyroptosis is a key cellular inflammatory response against extracellular pathogens like microbial and viral infections, ligands, or cellular perturbations [32]. However, pyroptosis is also known to play a critical role in various inflammatory diseases [33, 34]. Pyroptosis belongs to the innate immune response and can be classified as a cell death form orchestrated through canonical and/or non-canonical signaling pathways involving members of the gasdermin (GSDM) family.…”
