Molecular Beacon Probes of Oligonucleotides Photodamaged by Psoralen

Abstract: Ultraviolet A (UVA)-irradiated 4'-hydroxymethyl-4,5',8-trimethyl psoralen (HMT) in the presence of a poly-dT(17) and dA(7) TTA(8) oligonucleotides produces HMT-dT(17) and HMT-dA(7) TTA(8) adducts in aqueous solution. In this article, we determine whether these HMT-dT(17) and HMT-dA(7) TTA(8) adducts can be detected with a molecular beacon (MB) probe. We measure the degree of damage in dT(17) and dA(7) TTA(8) solutions containing UVA-activated HMT via monitoring the decrease in MB fluorescence. Photoproduct for… Show more

“…This light-activated insecticide is expected to become an effective alternative for pest control due to its lower toxicity to mammals, higher insecticidal efficiency, and environmental friendliness [ 11 , 12 , 13 ]. Previous studies showed that furanocoumarins activated by light could react with bases on DNA double-strands to form double-crosses and destroy DNA replication, inhibit the growth of a variety of cancer cells, and become a potential drug for treating cancer [ 14 ]. The effect of furanocoumarins on photoactivated pesticides was first shown by Berenbaum.…”

“…Thus, while LC‐MS/MS is necessary to determine a given population of adducts in addition to total binding level, DNA digestion to nucleosides and postdigestion purification make the procedure time‐consuming. Shire and Loppnow showed that oligonucleotides may be desorbed in matrix‐assisted laser desorption ionization (MALDI) by using the appropriate matrix and charge mode . In this report, we also tested whether surface‐enhanced laser desorption ionization (SELDI‐TOF) technology , a variant of MALDI which combines chromatography with mass spectrometry through the use of affinity surfaces, may be used for psoralen‐DNA adduct detection.…”
