Molecular Authentication of Alisma orientale by PCR-RFLP and ARMS

Abstract: As a widely used medicinal plant, Alisma orientale is always a possible target for fraudulent labeling. The internal transcribed spacer (ITS) regions of nuclear ribosomal DNA (nrDNA) of the six species of genus Alisma were sequenced, and two variant sites were found to be specific for A. orientale. Polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) analysis and amplification refractory mutation system (ARMS) analysis were applied to the ITS region for the identification of A. orienta… Show more

“…[9][10][11][12] ARMS (amplification refractory mutation system), which has also been described as allele-specific polymerase chain reaction (PCR), is on the basis of single nucleotide polymorphisms (SNPs) of different sequences and has been successfully used for herbs and their adulterants authentication. 5,13,14) gDNA SSH (genomic DNA suppression subtraction hybridization) is an efficient and sensitive PCR-based gDNA subtraction method and is capable to be applied to germplasm authentication, genetic mapping and gene tagging, molecular marker-assisted breeding and species identification. 15,16) In the present study, sequences of chloroplast genes (matK and rbcL), nuclear rDNA ITS regions, mitochondria gene nad1 intron2 would be analyzed and diagnostic primers for ARMS would be designed for population authentication.…”

“…PCR-RFLP is one of the most popular DNA-based techniques for species identification in biology, medicine, and food science 16 . The method was successfully used to differentiate Alisma orientale from Alisma species 17 and Bulbus fritillariae from its adulterants 18 and to identify species in anchovy pastes on the market 19 . The aim of the present study was to identify the antioxidant properties of the three 'Rang Chuet' species, including T. laurifolia, Crotalaria spectabilis, and Curcuma aff.…”
