Molecular Analysis of GmFAD3A in Two Soybean Populations Segregating for the fan, fap1, and fapnc Loci

Abstract: Soybeans [Glycine max (L.) Merr.] have undesirable levels of polyunsaturated fatty acids in their oil that result in oxidative instability and poor flavor. The process of hydrogénation improves the stability but creates undesirable trans fats. Lines carrying fan genes have decreased linolenic acid (18:3) content. Changes in transcription or activity of the desaturase encoded by the GmFAD3 gene cause a reduction in 18:3 content in certain lines.

“…FAD3A encodes a linoleate desaturase that introduces a third double bond in linoleic acid to form linolenic acid. Cardinal et al [ 65 ] suggest that lower expression of the FAD3A allele in N0304-303-3 is likely to cause a lower level of linolenic acid (18:3) content and is mirrored in our finding. M23 has a 164 kb deleted region containing the fatty acid desaturase gene FAD2-1A [ 66 , 67 ] and therefore, as expected, we detected little more than background expression of the FAD2-1A gene in M23.…”

“…In addition to the FAD3B and FAD3C genes, the FAD3A gene has a strong influence on the concentration of linolenic acid (Cardinal et al 2011). Different combinations of mutations in these three genes are capable of producing soybeans with low levels of linolenic acid (1%) .…”

“…N98-4445 is a F 3 -derived higholeate (563.1 g kg -1 ), low-linoleate (26.3 g kg -1 ;homozygous for fan PI 123440) line that is a sister line of the released germplasm N98-4445A developed by the USDA-ARS in Raleigh, NC (Burton et al 2006). Satelite is a lowpalmitate (39.8 g kg -1 ; homozygous for fap nc and fap1), low-linolenate cultivar (fan PI 123440) (Cardinal et al 2007;Bachlava et al 2008aBachlava et al , b, 2009bCardinal et al 2011). FAHH00 (F 4 derived lines) was derived from the cross of a high yielding line, 'Corsica' (Kenworthy 1996), and N97-3681-11, a low linolenic (fan PI 123440) and low palmitic acid (homozygous for fap nc and fap1) line.…”
