Molecular alterations of bone quality in sequesters of bisphosphonates-related osteonecrosis of the jaws

Abstract: The present data show that jaw bone quality can be altered with an overmineralization and ultrastructural modifications of apatitic mineral in bone sequesters of BP-related ONJ.

“…The Raman bands due to PMMA (a commonly used embedding resin when handling biological tissues) are ~1365 cm −1 (weak) and 1390 cm −1 (medium); thus, in all measurements obtained in embedded tissue, the PMMA contribution was spectrally subtracted, including the Raman images. We have previously utilized the 1375 cm −1 Raman band to describe PG concentration and distribution in bone, while the results presented in the current study show its utility in cartilage analysis.…”

“…Over the last decade several investigators have reported the usefulness of Raman spectroscopy in the diagnosis of a variety of bone diseases in humans, that has been summarized chronologically as a table in the supplementary information 14 – 22 . These reports range from studies looking at mineralization and structural deterioration of bone in osteoporosis 14 , 20 , structural deterioration of collagen of nails in osteoporosios 22 , monitoring treatment of osteoporosis 15 , 17 , and monitoring a major adverse reaction to the treatment of osteoporosis such as osteonecrosis of jaw 19 . Other diseases affecting the structure of bone such as osteomalacia 18 , and osteogenesis imperfecta 21 , 22 have also been studied using Raman spectroscopy.…”

“…[66][67][68] Some model and treatment studies have gone further to examine relationships between changes in osteoporotic bone composition with alternations in bone material properties. 52,[69][70][71][72] For example, the mineral/matrix ratio and indentation modulus were found to be reduced in cancellous bone from the lumbar spine of female donors with osteoporosis.…”
