This study evaluated the inhibitory effect of ethyl acetate fraction of Solanum macrocarpon L. leaves on cholinergic, monoaminergic, purinergic enzyme activities and identification of bioactive compounds. 2,2‐azino‐bis‐3‐ethylbenthiazoline‐6‐sulphonic acid (ABTS), metal ion chelating radicals scavenging activity, total phenolic and flavonoid contents were determined using standard methods. The inhibitory effects of the ethyl acetate fraction of S. macrocarpon on the specified enzymes were determined spectroscopically. Ethyl acetate fraction of S. macrocarpon showed a high ABTS radical scavenging activity and metal ion chelating radical scavenging activity. The fraction showed high contents of phenol and flavonoid. The fraction also inhibited AChE, BChE, MOA, tyrosinase, arginase, ecto‐5′ ‐nucleotidase, PDE‐5, ACE and stimulated the activity of Na+/K+‐ATPase. Bioactive compounds present in ethyl acetate fraction of S. macrocarpon leaves include kaempferol‐3‐rutinoside, rutin among others. Ethyl acetate fraction of S. macrocarpon leaves inhibited cholinergic, monoaminergic, and purinergic enzymes and had a stimulatory effect on Na+/K+‐ATPase. Practical applications Solanum macrocarpon L. leaves are commonly consumed as vegetable. The enormous medicinal potential of the ethyl acetate fraction is invaluable as it’s a probable source of polyphenols characterized with strong antioxidant activity. In addition, the leaves are used as functional foods and may play a crucial role in the management/treatment of diabetes mellitus, neurodegenerative diseases, and erectile dysfunction.

This study investigated effect of p-coumaric acid (PCA) on erectogenic enzyme activity and non-protein thiol level in the penile tissue of normal and doxorubicin (DOX)induced oxidative stress male rat. Sixty-four (64) adult male rats weighing between 170 and 180 g were used for this work. After 14 days of acclimatisation, the rats were divided into eight groups (n = 8). Rats were orally pre-treated with PCA dose dependently (50 and 100 mg/kg body weight [b.w.t]) and vitamin E (100 mg/kg b.w.t) for 14 days before induction with a single dose of DOX (15 mg/kg b.w.t, via i.p.). The result revealed that arginase, acetylcholinesterase (AChE), angiotensin-I-converting enzyme (ACE), phosphodiesterase-5 (PDE-5), adenosine monophosphohydrolase (AMPdase) activities were significantly (p < 0.05) higher in the DOX-induced rats as against the control, which was significantly p < 0.05) higher when compared to normal rats treated with PCA. PCA also improved non-protein thiol level in the penile tissue of both normal and DOX-induced rats. Hence, this study revealed that PCA is capable of causing inhibitory effects on the activities of enzymes, associated with oxidative stress-induced erectile dysfunction (ED) and could also be used as an aphrodisiac agent in the management/treatment of ED. Investigation into modulatory effects of local condiments produced from fermented legume (African locust bean and soybean) seeds on activities of enzymes relevant to endothelial function and cardiovascular disease (arginase, phosphodiesterase‐5, acetylcholinesterase, and, ecto 5′‐nucleotidase) in vitro was the focus of this study. The condiments were prepared according to traditional methods of fermentation. Thereafter, modulatory effects of aqueous extracts from the condiments on activities of the enzymes were subsequently carried out. Results showed the extracts significantly inhibited activities of arginase, phosphodiesterase‐5 and acetylcholinesterase, while the activity of ecto 5′‐nucleotidase was stimulated at sample concentrations tested. Thus, the observed enzyme modulatory properties exhibited by the condiments could be novel mechanisms to support their use as functional foods and nutraceuticals for the management of cardiovascular disease and associated endothelial dysfunction.
