Modulation of Neuronal Excitability by Serotonin-NMDA Interactions in Prefrontal Cortex

Abstract: Both serotonin and NMDA signaling in prefrontal cortex (PFC) are implicated in mental disorders, including depression and anxiety. To understand their potential contributions to PFC neuronal excitability, we examined the effect of co-activation of 5-HT and NMDA receptors on action potential firing elicited by depolarizing current injection in PFC pyramidal neurons. In the presence of NMDA, a low concentration of the 5-HT 1A agonist 8-OH-DPAT substantially reduced the number of spikes, and a low concentration o… Show more

“…These findings lead us to speculate that fear stress induces downregulation of postsynaptic 5-HT 1A receptors in the females and ultimately causing failure of 5-HT 1A receptor-mediated synaptic inhibition. The 5-HT 1A -N-methyl-d-aspartate (NMDA) receptor interaction is known to provide potential mechanisms underlying the 5-HTergic regulation in emotional processes (53,54). Therefore, their interaction may contribute to the different patterns of 5-HT 1A receptor-mediated synaptic responses.…”

“…Whole-cell current clamp techniques were used to measure action potential firing (30). During the recording of spontaneous firing, PFC slices were bathed in a modified ACSF with reduced Mg 2ϩ (0.5 mM) and slightly elevated K ϩ (3.5 mM) at room temperature.…”

“…5-HT2A receptors have also been reported to activate Akt and Erk (Watts, 1998; Johnson-Farley et al, 2005; Zhong et al, 2008), although the receptor-coupling mechanism of this regulation is not completely understood. 5-HT2A receptors also couple to the intracellular scaffolding protein β-arrestin2, which may have unique functions to initiate desensitization of 5-HT2A receptors and to direct ligand-selective signal transduction processes (Schmid et al, 2008; Schmid and Bohn, 2010).…”
