Modulation by Cell Trypsinization of Sendai Virus Expression in African Green Monkey Kidney Cells: First Infection and Establishment of a Carrier State.

Abstract: SUMMARYTwo African green monkey kidney (AGMK) cell lines, 37RC (interferonproducing) and Vero (non-interferon-producing), were infected by egg-grown Sendai virus passaged in eggs at high or low m.o.i. The appearance of haemadsorption, and cytopathic effect (c.p.e.) as well as the presence of haemagglutinating virions in the supernates were much more pronounced with a virus seed obtained with Io-a-diluted passages than with a seed obtained with undiluted inoculum. They were also independent of interferon produc… Show more

“…However, a peak of virus production was detected 48 to 72 hours after trypsinization of the cells. This indicates that the cells had become persistently infected (carriers) and were resistant to a reinfection (no increase in HAU or cell damage) by the parental egg-grown Sendai virus (7). As shown in Fig.…”

“…The presence of 1.2-epoxy-2-methylpropyl moiety instead of the isobutenyl group was suggested by the presence of a pair of methyl singlets at 8 1.33 and 1.30, and a one-proton doublet at 2.71 (J = 8.0). 7. Hydrogenations were carried out in ethanol containing catalytic amounts of platinum oxide and stirred for 1 hour in a hydrogen atmosphere.…”

“…The establishment and the culture technique of 37RC cells have been discussed in detail previously (Benedetto et aL, 1979). Briefly, the cells were grown in either T-25 Falcon flasks or in 24-well Linbro plates in Eagle's minimal essential medium (MEM) supplemented with 5 % foetal calf serum (Gibco) and antibiotics at 37 °C in a 5 % CO 2:95 % air, humidified atmosphere.…”
