Modifying alginate with early embryonic extracellular matrix, laminin, and hyaluronic acid for adipose tissue engineering

Abstract: Extracellular matrix provides both mechanistic and chemical cues that can influence cellular behaviors such as adhesion, migration, proliferation, and differentiation. In this study, a new material, HA-L-Alg, was synthesized by linking developmentally essential ECM constituents hyaluronic acid (HA) and laminin(L) to alginate (Alg). The fabrication of HA-L-Alg was confirmed by FTIR spectroscopy, and it was used to form 3D cell-carrying beads. HA-L-Alg beads had a steady rate of degradation and retained 63.25% o… Show more

“…The higher levels of collagen type IV, collagen type VI and laminin in the DAT may have had instructive effects in mediating adipogenic differentiation, as the basement membrane is known to play a fundamental role in adipogenesis (Kawaguchi et al, 1999;Uriel et al, 2008;Hsueh et al, 2017). Further supporting this possibility, chitosan and alginate composite hydrogels incorporating purified laminin or laminin-derived sequences have successfully enhanced the adipogenic differentiation of preadipocytes and ASCs in vitro (Chen et al, 2015(Chen et al, , 2016bHsueh et al, 2017).…”

“…One of the limitations of our experimental setup is that we cannot be sure about the complete integration of LN521 into the alginate structure at molecular level, but we speculate that some of the LN521 molecules were trapped in the internal network of the scaffold. The use of oxidizing agents as IO4when preparing the scaffolds can be addressed for future approaches, allowing interaction between aldehyde groups of the oxidized alginate and amine residues of the laminin, as previously reported [32].…”

“…The combination of adult stem cells from various sources within 3D polymeric structures has been one of the most approached to generate 3D adipose-like tissues. The majority of these works have been using 3T3-L1 [ 4 , 5 , 6 , 7 ], a murine pre-adipocyte cell line that has limited representation of human tissue physiology. In alternative, adult stem cells have been differentiated into the adipogenic lineage in a range of natural/synthetic materials [ 8 , 9 , 10 , 11 ].…”
