To examine the influence of adrenal secretions on neuroleptic induced catalepsy, we studied the effect of adrenocorticoids, noradrenaline (NA) or adrenaline (AD) on haloperidol (HAL) induced catalepsy in adrenalectomised (ADX) and sham-adrenalectomised (sham-ADX) rats. HAL (1 mg/kg, i.p.) induced a greater degree of catalepsy in ADX rats as compared to sham-ADX rats. Corticosterone (CORT, 1-2 mg/kg, s.c.) or dexamethasone (1-2 mg/kg, s.c.) attenuated the HAL catalepsy in sham-ADX but not in ADX rats. Further, when the HAL (1 mg/kg) catalepsy score was maximal (at 120 min), the rats were subjected to cold stress (3 degrees C for 10 min) or treated with NA, AD (2 micrograms/kg, i.v.) or CORT (2 mg/kg, s.c.). After cold stress procedure or CORT treatment, the catalepsy was significantly reduced in sham-ADX but not in ADX rats, whereas NA or AD infusion caused an immediate but short lasting significant decrease in HAL catalepsy in both sham-ADX and ADX rats. The anticataleptic effect of NA or glucocorticoids was blocked by an alpha 1 adrenoceptor blocker, prazosin. These findings suggest that peripheral noradrenergic and adrenergic mechanisms play an important role in the neuroleptic induced catalepsy. Such mechanisms may mediate the anticataleptic action of glucocorticoids.

Parkinson's disease (PD) is a consequence of specific progressive neurodegeneration of substantia nigra pars compacta dopaminergic neuron resulting depletion in Dopamine (DA). Ldopa is first line DA replacement therapy. Caffeine and Antimuscarinic agents are effective adjuvant in PD management. Caffeine is given to counteract the adverse effects of anticholinergics. Caffeine is considered as a very safe psychostimulant. Contrary, higher doses of Caffeine associated with tachycardia, nausea and vomiting. Anticholinergics at therapeutic doses produce amnesia and cognitive impairment, particularly intense in elderly with preexisting neurodegenerative disease. Little is known about association between caffeine use, toxicity and dependence and risk of common psychiatric and substance use disorder. In present study, Decaffeinated Tea Extract (DTE) has been screened for antimuscarnic and anticataleptic activity. The results indicate that DTE significantly inhibited Ach-induced contractions and Haloperidolinduced catalepsy. The effects are comparable to Atropine & Caffeine. As per our study findings, decaffeinated tea is an effective option as an adjuvant as compared to Caffeine and Atropine.

Herbal drugs are playing an important role in health care all over the world, especially in developing countries because they are being cheap, locally available and consider being safe. Neuroleptic drugs used in the treatment of schizophrenia and other affective disorders are known to produce extra pyramidal side effects. Hypericum hookerianum (Hh) is known to possess various therapeutic properties. We have earlier characterized its phytochemical and physic chemical properties. However, their role in modulating stress-induced central changes is unexplored. Thus, the present study was aimed to investigate the brain oxidative stress induced by haloperidol in swiss albino mice and evaluate the consequent changes in brain neurotransmitters levels such as Dopamine, serotonin, nor adrenaline and GABA. Results showed that Brain neurotransmitters in this study were decreased in schizophrenic mice which may be related to oxidative stress induced by haloperidol. This is reversed by the administration of ethanolic extract of Hypericum hookerianum (EEHh) which seems to be a highly promising herb in protecting the schizophrenicmice against oxidative damage and preventing brain complications such as reduction of neurotransmitters. Further studies are required to understand the mechanism of action of this herb and active principle responsible for this neuroprotection.
