Modification of decellularized vascular scaffold with conditioned medium to enhance cell reseeding

Abstract: Repopulation of decellularized vascular scaffolds (DVS) is limited because of change in the repertoire and ratios of the remaining extracellular matrix (ECM) proteins, for example, loss of glycoproteins and the retention of type I collagen. Pre-treatment of DVS with defined ECM proteins, which match the repertoire of integrin receptors expressed by the embryonic stem cells (mESCs) to be seeded, can increase the reseeding efficacy. mESCs mainly express high levels of functional receptors for LM and FN. Reseedin… Show more

“…14 Pre-treatment of decellularized vascular scaffolds with defined ECM proteins, which match the repertoire of integrin receptors expressed by the embryonic stem cells to be seeded, can increase the reseeding efficacy. 15 Modifying decellularized matrix derived from porcine small intestinal submucosa (SIS) with thrombomodulin alone enables robust activated protein C generation, and thrombomodulin activity is maintained after prolonged exposure to fluid shear and blood plasma. Heparin-modified SIS has a potent anticoagulant activity.…”

“…Pre-treating distributed switches with ECM proteins matching the integrin receptor library expressed by the seed cells can improve reimplantation. 79 A major problem in cell regeneration experiments is the black-box approach, which is the method used in most studies and the subsequent evaluation of different outcome parameters. The biomechanical aspects of the cell re-formation process have not been considered, such as whether parenchymal cells cross the vascular basement membrane during cell re-formation and how the ECM is damaged during direct injection, and the spatial and temporal resolution of the kinetics of recellularization is still poorly known.…”
