Modeling the intracellular dynamics for Vif-APO mediated HIV-1 virus infection

Abstract: The viral infectivity factor (Vif) was found to be essential for controlling HIV-1 virus infectivity. It targets cellular antiviral proteins in APOBEC family (APO) to trigger its fast degradation and inhibits APO packaging into nascent virion. In the present study, we propose a mathematical model to quantitatively study the intracellular dynamics of these typical virus-host interactions. Four sets of published experimental data were compared with simulation results to justify the model. Systematic parameter se… Show more

“…The model incorporates three existing ODE (ordinary differential equation) models of HIV-1 intracellular kinetics: Kim & Yin 29 , Reddy & Yin 18 and Wang & LuHua 30 . HIV has been studied in detail and all three of these models are based on a wealth of experimental data that characterizes the rates of HIV mRNA transcription, ratios of HIV protein translation and half-lives, protein cytoplasmic concentrations and interactions with host proteins, and the mechanisms and kinetics of viral replication and virion budding.…”

“…Wang and Lai (14) modeled the transcription of viral mRNA, translation of Gag, Tat, and the accessory protein Vif, the creation of Vif-Gag, Vif-APO, and Gag-APO complexes, where APO is a cellular antiviral protein, together with incorporation of these complexes into budding virions. The model was verified using several experimental data sets.…”

“…The results indicate that the detection method can successfully detect most particles in an image with accuracy of 98.61% and 97.65% for adaptor and clathrin images, respectively, and that the tracking algorithm can resolve merging and splitting issues encountered when analyzing dynamic, live-cell images of clathrin assemblies. Recently, much attention has been focused on the development of segmentation and particle tracking techniques for the analysis of large volumes of microscopy fluorescence images acquired during cellular level cell imaging [1,2]. Particle tracking is one method of acquiring information about cellular dynamics.…”
