Modeling of pneumococcal serogroup 10 capsular polysaccharide molecular conformations provides insight into epitopes and observed cross-reactivity

Abstract: Streptococcus pneumoniae is an encapsulated gram-negative bacterium and a significant human pathogen. The capsular polysaccharide (CPS) is essential for virulence and a target antigen for vaccines. Although widespread introduction of pneumococcal conjugate vaccines (PCVs) has significantly reduced disease, the prevalence of non-vaccine serotypes has increased. On the basis of the CPS, S. pneumoniae serogroup 10 comprises four main serotypes 10A, 10B, 10C, and 10F; as well as the recently identified 10D. As it … Show more

“…The determination of the glycosidic linkages was achieved by GLC-MS analysis of the partially methylated alditol acetates which identified: 4,5-Rib-ol, 3-Glc p , 4-Gal p or 5-Gal f , 3-Gal p , and 3-GalNAc p in the molar ratio of 0.4:1.6:2.1:1.0:0.7. These data are consistent with a linear hexasaccharide repeat unit without branch points and side chains, unlike the repeat units reported for related serotypes 10B ( 42 ) and 33B ( 43 ).…”

“…Like 33B, 33G and other serotypes in serogroup 33 (33C and 33D) also possess a hexasaccharide backbone containing ribitol-5-P. In addition, 33G has an O-acetylated β-Gal f residue, found in all members of serogroup 33 and lacking in serogroup 10 capsules (39, 40). The presence of O-acetyl groups in the 33G capsule is likely to have important biological relevance given that O-acetylation impacts the structure of the polysaccharide, and is an epitope in pneumococcal capsules readily recognised by the immune system (44, 49).…”

“…Cross-reactivity between serotypes Spn-10A, Spn-10B, Spn-10C and Spn-10F is complex with unclear structural reasons, serotype-specific rabbit antisera generated against a serotype recognized other serotypes, albeit with less affinity [ 67 ]. Ravenscroft and Kuttel recently published a molecular modeling study that together with relevant serological studies supports the inclusion of serotype 10A in a vaccine to best protect against serotypes 10 disease [ 68 ].…”
