When reactive oxygen species (ROS) accumulate in the body, they can lead to inflammatory bowel disease (IBD) through their oxidative damages to DNA, proteins, and lipids. In this study, a thermosensitive hydrogel-based nanozyme was developed to treat IBD. We first synthesized a manganese oxide (Mn3O4) nanozyme with multienzyme activity followed by physically loading with a thermosensitive hydrogel poly(d,l-lactide)-poly(ethylene glycol)-poly(d,l-lactide)-based triblock copolymer (PDLLA-PEG-PDLLA). Then, a mouse model based on the inducement of dextran sulfate sodium (DSS) was built to assess the ROS targeting, scavenging, as well as anti-inflammatory ability of Mn3O4 nanozymes-loaded PDLLA-PEG-PDLLA (MLPPP). Because of the sharp gelation behavior of PDLLA-PEG-PDLLA in body temperature, the MLPPP nanozyme can easily target the inflamed colon after colorectal administration. Following the formation of a physical protection barrier and sustained release of manganese oxide nanozymes that had diverse enzymatic activities and can effectively scavenge ROS, the administration of the MLPPP nanozyme had a high efficacy for treating colitis mice; importantly, after the treatment with this novel nanoformulation, the levels of the pathological indicators in colons as well as in sera of colitis mice were even comparable to healthy mice. Therefore, the MLPPP nanozyme has a potential application for nanotherapy of IBD and would have great clinical translation prospects.

The accumulation of reactive oxygen species (ROS) and minimal osteogenic raw material in the osteoporotic bone microenvironment greatly inhibits the activity of osteoblasts. Herein, it is originally proposed to construct a biomatrix multifaceted bone microenvironment amendment ‐Mineralized zippered G4‐Hemin DNAzyme hydrogel (MDH)‐to improve osteoporotic osteogenic capacity and promote high‐quality bone defect repair. The programmed design of the rolling circle amplified DNA hydrogel synthesis system allows the introduction of massive amounts of zippered G4‐Hemin DNAzyme in MDH. The zippered G4‐Hemin DNAzyme highly mimics the tight catalytic configuration of horseradish peroxidase and exerts excellent enzyme‐like activity with considerable ROS molecule scavenging ability. In addition, the DNA amplification by‐product pyrophosphate is ingeniously employed as a sufficient phosphorus source, thus constituting an autonomous mineralization system for waste reuse through the introduction of pyrophosphate hydrolase and calcium ions, which deposits in MDH as an osteogenic raw material and addresses the challenge of DNA hydrogel bio‐application stability. The remarkable in vitro and in vivo outcomes demonstrate that MDH can effectively improve the oxidative stress status of osteoblasts, restore the balance of mitochondrial membrane potential, and reduce apoptosis, ultimately demonstrating superior osteogenic capacity.

Possessing the largest surface area of mucosa in the body, the gastrointestinal (GI) tract can easily sufferfrom inflammatory damage under various adverse external exposures, resulting in the occurrence of inflammatory bowel disease (IBD). Excessive reactive oxygen species (ROS) usually lead to local mucosal injury, accelerate the formation of niduses, andamplify the inflammatory and immune response. Antioxidant therapy, therefore, is considered as a potential strategy against IBDs. Herein, a series of novel dihydromyricetin‐based nanoassemblies with excellent antioxidant activities and high dispersion in GI tract as oral preparations are developed for the targeted IBD treatment. By changing raw materials, the current strategy can be well extended to the preparation of other insoluble natural flavonoid compound‐based nanoassemblies. The well‐designed dihydromyricetin‐PEG 1000‐based nanoassemblies (DMY‐1000 NAs) with high stability and great ROS scavenging capacity in the harsh environment of GI tract hold an admirable targeted capability toward the intestinal inflamed lesions. Therefore, these biocompatible DMY‐1000 NAs show promising therapeutic effects for typical IBDs including ulcerative colitis and Crohn's disease in murine models. This study not only provides a new method for constructing antioxidant therapy platforms but also illustrates their prominent therapeutic promise against IBDs.
