MKK3- and MKK6-Regulated Gene Expression Is Mediated by the p38 Mitogen-Activated Protein Kinase Signal Transduction Pathway

Abstract: The p38 mitogen-activated protein (MAP) kinase signal transduction pathway is activated by proinflammatory cytokines and environmental stress. The detection of p38 MAP kinase in the nucleus of activated cells suggests that p38 MAP kinase can mediate signaling to the nucleus. To test this hypothesis, we constructed expression vectors for activated MKK3 and MKK6, two MAP kinase kinases that phosphorylate and activate p38 MAP kinase. Expression of activated MKK3 and MKK6 in cultured cells caused a selective incre… Show more

“…48,49 Similarly, the expression of activated MKK3 promoted Bim EL phosphorylation, 47 but promoted activation of JNK as well as p38. Since MKK3 is established as a selective activator of p38, 50 this suggests that the assay conditions may not have been stringent. These examples highlight some of the concerns that arise when using transient overexpression systems to address issues of sufficiency.…”

“…The dual speci®city kinase that phosphorylate ERK5 is known as MEK5 or MKK5 (Zhou et al, 1995;. The recently identi®ed dual speci®city kinase that speci®cally involved in the regulation of p38MAPK is known as MKK6 (Raingeaud et al, 1996;Moriguchi et al, 1996;Han et al, 1996;Stein et al, 1996). Likewise, newly characterized dual specificity kinase that speci®cally activates JNK through phosphorylation is named MKK7 Yao et al, 1997;Tournier et al, 1997;Wu et al, 1997).…”

“…Both, MAPKs and SAPKs are regulated by upstream kinases (Raman et al, 2007) and consist of a three module system transducing the signal through subsequent phosphorylation into the nucleus. Two kinases, MKK3 and MKK6, are known to activate p38 (Raingeaud et al, 1996;Schaeffer and Weber, 1999).…”
