Mitotic protein kinase-driven crosstalk of machineries for mitosis and metastasis

Abstract: Accumulating evidence indicates that mitotic protein kinases are involved in metastatic migration as well as tumorigenesis. Protein kinases and cytoskeletal proteins play a role in the efficient release of metastatic cells from a tumor mass in the tumor microenvironment, in addition to playing roles in mitosis. Mitotic protein kinases, including Polo-like kinase 1 (PLK1) and Aurora kinases, have been shown to be involved in metastasis in addition to cell proliferation and tumorigenesis, depending on the phosph… Show more

“…During mitosis, numerous proteins are phosphorylated by kinases, such as cyclin-dependent kinase 1 (CDK1) and CPC (Dephoure et al, 2008; Enserink and Kolodner, 2010; Kim et al, 2022; Salaun et al, 2008; Vagnarelli, 2021). Although the key kinases involved in phosphorylation have been well researched, the role of the phosphorylated proteins in mitosis remains unknown.…”

“…The top five derived hub genes were PLK1, KIF20A, RRM2, CDC20, and UBE2C in M1CM groups and RRM2, CDC20, CCNB1, PLK1, and UBE2C in M2CM groups. Polo like kinase 1 (PLK 1) plays an important role in the regulation of the cytoskeleton network during mitosis ( 32 ), and li et al. demonstrated PLK1 expression was decreased in NPCs of degenerative IVDs ( 33 ).…”

“…Indeed, NFκB inhibition by PAs correlates with reduction of MMP-9 and reduced migration [ 105 ]. Interestingly, novel targets of PAs, Aurora A and B kinases, are also involved in mediating EMT via activation of NFκB [ 95 , 106 ]. However, to date, no in vitro studies have included the role of PAs in the crosstalk of Aurora A/B and NFκB.…”
