Aim Phylogeographical structure in the black salamander (Aneides flavipunctatus) was inferred using two independent genetic datasets. Concordance between the datasets was sought in order to evaluate earlier suggestions of species‐level breaks and evidence of vicariance and long‐term isolation within the complex. We hypothesized that major phylogeographical breaks would either correspond to current tectonic plate boundaries or to historical geological processes. Location North‐western California and southern Oregon (USA). Methods Three mitochondrial DNA (mtDNA) genes were sequenced for 240 black salamanders from 136 localities, and up to 13 nuclear DNA loci were sequenced for 145 black salamanders representing 93 localities. Phylogenetic analysis of our mitochondrial dataset was performed to recover major lineages, while spatial clustering analysis of our nuclear dataset was utilized to identify points of concordance with our mtDNA phylogeny. Levels of gene flow were estimated for all contact zones. Results Our mitochondrial phylogeny recovered four major lineages. Cluster analysis of our nuclear dataset is consistent with a four‐population scheme, with the boundaries matching those of the mtDNA lineages. Gene flow across a contact zone in southern Humboldt between two of the populational units is extremely limited (2Nm < 1). In what is identified as the Central Core population, two distinctive subpopulations were delineated based on nuclear data, but mitochondrial data are discordant. Main conclusions The Aneides flavipunctatus complex comprises at least four species‐level units. Two of the boundaries between these units are associated with current tectonic plate boundaries. The contact zone between our Northwest and Central Core populations lies adjacent to the Mendocino Triple Junction (MTJ), where the Humboldt and North American plates meet, while the area separating the Santa Cruz and Central Core populations corresponds to the boundary between the Pacific and Humboldt tectonic plates. The phylogeographical break within the Central Core population lies in a region in which uplift occurred that is associated with the historical position of the migrating MTJ.

Geographic and genetic isolation in spring-associated Eurycea salamanders endemic to the Edwards Plateau region of Texas

Populations of neotenic, spring-associated salamanders of the genus Eurycea occupy discontinuous sites throughout the Edwards Plateau of central Texas and many warrant conservation attention. Here we used DNA sequence data from a nuclear (rag1) and a mitochondrial (ND4) gene to determine (1) the extent of genetic isolation among seven Edwards Plateau Eurycea populations and (2) the relationship between genetic distance and both geographic distance and hydrogeological features. Coalescentbased methods detected little gene flow among the sampled Eurycea populations, and we were unable to reject a model of complete isolation for any pair of populations. These findings were consistent with the relatively high genetic distances we detected among the sampled Eurycea populations (pairwise / ST ranged from 0.249 to 0.924). We detected a positive correlation between genetic distance and geographic distance, which is consistent with a pattern of isolation by distance. However, while controlling for geographic distance, we did not detect a positive relationship between genetic distance and aquifer or river distance. Thus, we found no evidence that aquifers and/or rivers serve as dispersal corridors among isolated Eurycea populations. Based on these results, we have no evidence that re-colonization of spring sites by migrant salamanders following local extirpation would be likely. Our findings indicate that spring-associated Eurycea salamander populations occupying the Edwards Plateau region are genetically isolated, and that each of these populations should be considered a distinct management unit.

The Pacific iguanas of the Fijian and Tongan archipelagos are a biogeographic enigma in that their closest relatives are found only in the New World. They currently comprise two genera and four species of extinct and extant taxa. The two extant species, Brachylophus fasciatus from Fiji, Tonga, and Vanuatu and Brachylophus vitiensis from western Fiji, are of considerable conservation concern with B. vitiensis listed as critically endangered. A recent molecular study has shown that Brachylophus comprised three evolutionarily significant units. To test these conclusions and to reevaluate the phylogenetic and biogeographic relationships within Brachylophus, we generated an mtDNA dataset consisting of 1462 base pairs for 61 individuals from 13 islands, representing both Brachylophus species. Unweighted parsimony analyses and Bayesian analyses produced a well-resolved phylogenetic hypothesis supported by high bootstrap values and posterior probabilities within Brachylophus. Our data reject the monophyly of specimens previously believed to comprise B. fasciatus. Instead, our data demonstrate that living Brachylophus comprise three robust and well-supported clades that do not correspond to current taxonomy. One of these clades comprises B. fasciatus from the Lau group of Fiji and Tonga (type locality for B. fasciatus), while a second comprises putative B. fasciatus from the central regions of Fiji, which we refer to here as B. n. sp. Animals in this clade form the sister group to B. vitiensis rather than other B. fasciatus. We herein describe this clade as a new species of Brachylophus based on molecular and morphological data. With only one exception, every island is home to one or more unique haplotypes. We discuss alternative biogeographic hypotheses to explain their distribution in the Pacific and the difficulties of distinguishing these. Together, our molecular and taxonomic results have important implications for future conservation initiatives for the Pacific iguanas.
