The intratracheal administration of interleukin (IL)-13 to hyperresponsive BP2 mice induces bronchopulmonary hyperreactivity (BHR), eosinophilia, mucus and MUC5AC accumulation, similar to those observed after ovalbumin (Ova) treatment when mice are immunized. mRNAs for IL-4 peaked at 6 h after Ova challenge, then vaned, whereas IL-13 expression was stable for a longer period, suggesting different effects. Inhalation of aerosolized methacholine by immunized mice 72-96 h after Ova reduced epithelial mucus content, and enriched the bronchoalveolar lavage fluid (BALF) mucus. The role of granulocytes for mucus accumulation was studied using vinblastine or the antigranulocyte antibody RB6-8C5, which interfered to a limited extent only with allergen-induced mucus accumulation. By contrast, eosinophilic and neutrophilic inflammation, as well as BHR, were completely suppressed. Granulocytes are thus involved in Ova-induced BHR, whereas mucus accumulation and BHR are unrelated. Granulocytes seem to be more implicated in rmIL-13-induced mucus, which is reduced by the antigranulocyte antibody, whereas BHR is unaffected. The glucocorticosteroid dexamethasone reduced all the parameters evaluated after Ova or after rmIL-13. Because the effects of IL-13 are glucocorticoid-sensitive, they probably involve secondary mechanisms.

In a crossover study, volunteers consumed a diet supplemented with either wheat or rye bread. Levels of faecal mucus were determined and the adhesion of five selected probiotics to immobilized intestinal mucus, was assessed. No significant difference in the concentration of faecal mucus or adhesion of the probiotics to the mucus was observed during the two dietary periods. However, male subjects were observed to excrete significantly more mucus during the rye bread supplemented diet. From these observations it was concluded that these two diets, with a low or high fibre content, did not change the potential binding sites for the tested probiotics or affect the level of mucus excreted in the faeces, however, on a high fibre diet men excrete more faecal mucus.

Study on the effects of laminarin, a polysaccharide from seaweed, on gut characteristics

This study investigates whether laminarin (β1-3,β1-6-glucan), a polysaccharide from seaweed, exhibits beneficial properties for human health by analysing its effects on intestinal parameters. Anaerobic batch culture fermenters were used for the screening of the in vitro utilization of laminarin by the human gut microflora through the monitoring of biochemical and microbiological parameters. Additionally, the influence of laminarin ingestion on the composition of intestinal mucus (neutral mucins, sialomucins and sulphomucins) was studied in rats. Laminarin was almost totally (more than 90% used) fermented after 24 h of incubation with human intestinal bacteria. It was not selectively used by bifidobacteria and lactobacilli, but increased the production of propionate and butyrate. Variations of mucus composition were observed in jejunum, ileum, caecum and colon, both in lumen content and in intestinal wall, of rats after ingestion of this polysaccharide. Due to its effects on mucus composition, laminarin could influence the adherence and the translocation of bacteria across the epithelial wall. In conclusion, laminarin seems to be a modulator of the intestinal metabolism by its effects on mucus composition, intestinal pH and short chain fatty acid (SCFA) production, especially butyrate.
