miRNA Profiling for Diagnosis, Prognosis and Stratification of Cancer Treatment in Cholangiocarcinoma

Abstract: Cholangiocarcinoma (CCA) is a lethal malignancy originating from the biliary tract epithelium. Most patients are diagnosed at an advanced stage. Even after resection with curative intent, prognosis remains poor. Previous studies have reported the evolving role of miRNAs as novel biomarkers in cancer diagnosis, prognostication and chemotherapy response. Various miRNAs, such as miR-21, miR-26, miR-122 and miR-150, have been identified as possible blood-based biomarkers for noninvasive diagnosis of CCA. Moreover,… Show more

“…MicroRNAs (miRNAs/miRs), a class of non-coding RNA molecules consisting of 18-25 nucleotides, have been verified to regulate the translation of various target genes and are involved in cancer occurrence, metastasis, progression and recurrence (5,6). In ccA, certain miRNAs have been previously demonstrated to exert stimulatory/suppressive effects on cancer progression (7). For example, miR-383 was found to act as a tumor suppressor in ccA and inhibits tumor development through decreasing the expression of interferon regulatory factor 1 (8).…”

“…Currently, prognostic microRNA (miRNA) expression signatures in various cancers, such as colon cancer (7), clear cell renal cell carcinoma (8) and cervical cancer (9) have attracted the attention of researchers. Therefore, the miRNA signature has been regarded as an important change in cholangiocarcinoma progression and therapy (10).…”

“…CCA has a low prevalence, but a high mortality rate and a poor 5-year survival rate (20). CCA often develops chemotherapeutic resistance and, as a result, the only current curative treatment is surgical resection (21). The majority of patients are with CCA diagnosed in advanced stages due to the nonspecific presentation of CCA, and surgical resection may not be suitable for these patients (22).…”
