miRNA‐216 knockdown has effects to suppress osteosarcoma via stimulating PTEN

Abstract: The aim of this study is to explain the effects and mechanism of miRNA‐216 in osteosarcoma. We firstly evaluated the PTEN expression in 30 pairs of tumor and adjacent tissues which were from the 30 osteosarcoma patients. In the following cell experiments, we measured the cell proliferation, cell cycle, cell invasion, and migration abilities of NC (normal control) group, BL (blank) group, siRNA (miRNA‐216 inhibitor) group, and siRNA+PTEN inhibitor group. Furthermore, we measured the relative protein expression … Show more

“…Numerous recent studies have demonstrated the aberrant miRNA expressions in osteosarcoma, which contribute to various aspects of osteosarcoma, including proliferation, differentiation, migration, and apoptosis (Jiang et al, 2020; Liang et al, 2013; Liu et al, 2020; Luo et al, 2020). The miR‐181a is an important miRNA with oncogenic function in osteosarcoma.…”

“…miRNA-216 knockdown promotes apoptosis through PTEN/PI3K/AKT and related downstream genes P53 and MMP-2/9, thus exerting antitumor effects. 104 High expression of miRNA-1226-3p was associated with lower overall survival. It can inhibit the proliferation, migration, and invasion of osteosarcoma cells by targeting TRIAP3 and promote the apoptosis of osteosarcoma cells.…”

“… 101 miRNA-21, miRNA-216, miRNA-155, and miRNA-524 can enhance the proliferation of osteosarcoma cells by targeting PTEN to activate the PI3K/Akt signaling pathway. 102 105 The PI3K/AKT signaling pathway is unregulated in most localized diseases and in 100% of advanced diseases, implying that alterations in this pathway may be a prerequisite for inhibiting osteosarcoma progression. 106…”

“…For example, knockdown of miR-216 suppresses the progression of osteosarcoma by targeting PTEN. 19 MiR-183-5p attenuates cerebral ischemia injury by negatively regulating PTEN. 9 In this study, we found that miR-195-5p was significantly downregulated during cerebral I/R injury.…”
