Background Previous reports showed that APOC1 was associated with several cancers but the function of APOC1 in cervical cancer was unknown. This study aimed to investigate the clinical effect and function of APOC1 in cervical cancer. Materials and Methods In this study, the relative expression of APOC1 in cervical cancer was detected by RT-qPCR. In order to determine the cell proliferation and migration and invading ability and apoptosis more accurately, we used CCK8 assay, Edu assay, wound healing assay, migration and invasion assay, flow cytometry assay, co-immunoprecipitation, proteomics and Western blot by silencing and overexpressing APOC1 , respectively. The role of APOC1 on tumor progression was explored in vitro and vivo. Results The relative expression of APOC1 in cervical cancer tissues was up-regulated (P<0.05). In cervical cancer cell lines, silencing of APOC1 restrained cell progression and EMT, while over-expression of APOC1 accelerated cell progression and EMT in vivo and vitro (P<0.05). Conclusion APOC1 acts as an oncogene in cervical cancers and knockdown of APOC1 inhibited cervical cancer cells growth in vitro and in vivo. There is a close relationship between the relative expression of APOC1 and clinical outcome in cervical cancer patients.

A prognostic five immune-related long noncoding RNA signature for patients with colon cancer

Background: Our study aimed to identify immune related long non-coding RNAs (LncRNAs) to serve as potential prognostic indicators and immune therapeutic targets in patients with colon cancer.Methods: The Cancer Genome Atlas (TCGA) and Molecular Signatures Databases (MSigDB) database were used to identify immune related lncRNAs in patients with colon cancer. The least absolute shrinkage and selection operator (LASSO) analysis, and multivariate Cox proportional hazards regression analysis were employed to screen prognostic lncRNAs and construct immune-related multi-lncRNA signature. We used time-dependent receiver operating characteristic curve to assess the performance of this signature in colon cancer by calculating the area under the curve (AUC). Univariate and multivariate Cox regression analysis were performed to verify the independence of the prognostic value of this signature in colon cancer.Results: Five immune related lncRNAs (AC025575.2, AL161729.4, ELFN1-AS1, LINC00513, MIR210HG) were found to be significantly associated with overall survival (OS) of patients with colon cancer. Then, we developed a five immune-related lncRNA signature. According to this signature, patients were ranked into a high risk group (n = 208) and a low risk group (n = 209). Kaplan-Meier curve and log-rank method showed that patients in high risk group had worse OS than patients in low risk group (P = 5.5644e-05). AUC for predicting 3 year survival and 5 year survival was 0.776 and 0.762 respectively, which indicated good performance of this signature. Finally, this five immune-related lncRNA signature was demonstrated to be independently associated with prognosis of patients with colon cancer.Conclusion: We developed a five immune-related lncRNA signature as a prognostic biomarker for patients with colon cancer.

Differential Expression Profiles of Long Noncoding RNA and mRNA in Dexamethasone-Induced Apoptosis of Human Bone Marrow Mesenchymal Stem Cells

Background Abnormalities in apoptosis, cell cycle, and proliferation of human bone marrow mesenchymal stem cells (hBMSCs) significantly impact bone metabolism and remodeling, and thereby cause various skeletal disorders. Long-term exposure to a high dosage of dexamethasone (Dex) induces apoptosis and inhibits proliferation of mesenchymal stromal cells (MSCs), which are probably the primary causes of osteoporosis (OP) and steroid-induced osteonecrosis of the femoral head (SONFH). However, to date, the exact mechanisms of Dex-induced apoptosis of BMSCs are still poorly defined. Methods A microarray was used to identify differentially expressed lncRNA and mRNA in Dex-induced apoptosis of hBMSCs, and bioinformatics was used to further explore the role of these differentially expressed lncRNAs and mRNAs by the coding and noncoding (CNC) network. Furthermore, validation of the microarray results was performed by quantitative real-time PCR (qRT-PCR) analysis. Results The microarray analysis identified a total of 137 differentially expressed mRNA (90 up-regulated and 47 down-regulated) and 90 differentially expressed lncRNA (61 up-regulated and 29 down-regulated) in Dex-induced apoptosis of hBMSCs. The differentially expressed mRNA and lncRNA were associated with the regulation of cell apoptosis. Meanwhile, several signaling pathways involved in the regulation of cell apoptosis, including mTOR signaling pathway, Ras signaling pathway, HIF-1 signaling pathway, NF-kappa B signaling pathway, and TGF-beta signaling pathway, also were identified in interaction net of the significant pathways (Path-Net) analysis. Furthermore, the CNC network further identified 78 core regulatory genes involved in the regulation of apoptosis. Besides, validation by qRT-PCR of the key differentially expressed mRNA and lncRNA, reported to be closely related to cell apoptosis, confirmed the reliability of the microarray dataset. Conclusions Collectively, we utilized microarray to identify differentially expressed lncRNA and mRNA in Dex-induced apoptotic hBMSCs, and bioinformatics to explore the interaction between the differentially expressed genes. This study demonstrates the molecular mechanisms of Dex-induced apoptosis of hBMSCs and provides a new research direction for the study of the pathogenesis of steroid-induced osteonecrosis of femoral head.
