miR-217 suppresses proliferation, migration, and invasion promoting apoptosis via targeting MTDH in hepatocellular carcinoma

Abstract: Hepatocellular carcinoma (HCC) has frequent incidence and the third highest mortality rate among cancers in the world. This study aimed to clarify the roles of miR-217 and metadherin (MTDH) in HCC. First, we identified that miR-217 expression was downregulated and MTDH expression was upregulated in the HCC tissues. Functional studies revealed that miR-217 negatively regulated MTDH expression via binding to the 3'-untranslated region of MTDH mRNA in the HCC cells. In our further studies, the miR-217 overexpress… Show more

“…The MTDH increases cell survival by promoting survival pathways such as PI3K and NF‐κB. Regulation of cell proliferation and metastasis via MTDH‐mediated autophagy is mediated by several miRNA, such as miR‐1297, miR‐145, miR‐98, and miR‐217. Our studies further revealed that miR‐22 increased the chemosensitivity through the miR‐22/MTDH/autophagy regulatory loop.…”

“…The EMT has an important role in tumor cell migration and invasion, and MTDH has been reported to be a contributor to EMT (27). In our study, we showed that MTDH is modulated by SNHG1; therefore, we speculated that SNHG1 also participates in the EMT process.…”

“…miRNAs are involved in regulation of the cell cycle and proliferation of HCC cells 49 , 50 . We previously reported that, for arsenite-transformed HaCat cells, circRNA_100284 regulates the cell cycle by acting as a sponge of miR-217 15 , a tumor suppressor that regulates cell proliferation in various cancers, including HCCs 51 . miR-217 inhibits cell proliferation through inducing arrest in the G2/M phase of the cell cycle 52 .…”
