miR-196b-Mediated Translation Regulation of Mouse Insulin2 via the 5′UTR

Abstract: The 5′ and the 3′ untranslated regions (UTR) of the insulin genes are very well conserved across species. Although microRNAs (miRNAs) are known to regulate insulin secretion process, direct regulation of insulin biosynthesis by miRNA has not been reported. Here, we show that mouse microRNA miR-196b can specifically target the 5′UTR of the long insulin2 splice isoform. Using reporter assays we show that miR-196b specifically increases the translation of the reporter protein luciferase. We further show that this… Show more

“…Among the latter steps, the regulation of translation is particularly important in cellular processes that require rapid changes in protein expression patterns. The translational efficiency of mRNAs is altered by RNA-binding proteins (RBPs) and noncoding (nc)RNAs such as microRNAs (Panda et al , 2014a and 2014b; Abdelmohsen et al , 2014). The impact of factors that regulate selective mRNA translation is a critical question in RNA biology.…”

“…Based on the recently developed crosslinking, ligation, and sequencing of hybrids technique (CLASH) (Helwak et al , ), they account for 3.8% of all miRNA interactions. In addition to the miRNA‐mediated posttranscriptional repression (Lee et al , ; Grey et al , ; Moretti et al , ; Zhou & Rigoutsos, ), direct target up‐regulation through 5′UTR interaction has been reported (Tsai et al , ; Liu et al , ; Panda et al , ). Along with translational, a transcriptional regulation by miRNA through 5′UTR has been also observed (Liu et al , ).…”

“…miR-483-3p has been also associated with the capacity of adipocytes to store lipids and dysregulation of its expression could directly inhibit adipose tissue expandability and lipid storage, which in turn affect other tissues by stimulating ectopic triglyceride storage and lipotoxicity [36]. Regarding the other miRNAs and their function, miR-196b has been associated with inflammatory bowel disease [37], insulin [38] and cancer [39]; miR-346 has been implicated in rheumatoid arthritis [40], osteogenic differentiation [41] and cancer [42]; miR-1247 was implicated in cancer [43,44]; miR135a was implicated in proliferation [45], inflammation [46] and calcification [47]; and miR-193a-5p was associated with cancer [48,49]. Although their effects in the intestine need to be evaluated, it is clear that HT could specifically modulate their expression and exert biological effects through this mechanism.…”
