MiR-155 Inhibitor-Laden Exosomes Reverse Resistance to Cisplatin in a 3D Tumor Spheroid and Xenograft Model of Oral Cancer

Abstract: Cisplatin resistance is one of the major concerns in the treatment of oral squamous cell carcinoma (OSCC). Accumulating evidence suggests microRNA (miRNA) dysregulation as one of the mediators of chemoresistance. Toward this, our previous study revealed the role of exosomal microRNA-155 (miR-155) in cisplatin resistance via downregulation of FOXO3a, a direct target of miR-155, and induction of epithelial-to-mesenchymal transition in OSCC. In the present study, we demonstrate the therapeutic potential of miR-15… Show more

“…The loading of the miRNA into exosomes was enhanced using this TAT-TAR interaction [ 42 , 43 ]. The modified calcium chloride (CaCl 2 ) method (CaCl 2 -heat shock) has successfully loaded nucleic acids into exosomes through forming CaCl 2 -nucleic acid complex, which was absorbed by exosomes under heat shock at 42 °C [ 44 46 ]. In addition, plasmid-mediated therapeutic molecule transfer has been gradually applied.…”

“…miR-155 is overexpressed in HypoExos from glioma [ 219 ] and hepatocellular carcinoma [ 19 ] and hypoxic TAMs [ 220 ] and is associated with cisplatin resistance [ 221 ]. miR-155 inhibitor-laden exosomes can reverse cisplatin resistance by suppressing drug efflux transporter protein expression in cisplatin-resistant OSCC [ 221 ], but whether HypoExo miR-155 can induce drug resistance by upregulating drug efflux is unknown. Single pathways are also utilized by HypoExos to promote drug resistance.…”

“…The cisplatin-treated cancers are attenuated with the suppression of tumorigenesis and metastases [ 2 ]. However, the biological applications of cisplatin are limited to eliminating the cancers because the average morbidity and mortality statistics are high [ 3 ], which may be due to prolonged usage that influence tumour adaptability [ 4 ] drug efflux [ 5 ] or acclimatisation in various biochemical [ 6 ] and immune networks [ 7 ] that modulate cancer -induced pathophysiology with widespread toxicity in the organs [ 8 ].…”
