Minireview: Nuclear Receptors, Hematopoiesis, and Stem Cells

Abstract: Nuclear receptors (NRs) regulate a panoply of biological processes, including the function and development of cells within the hematopoietic and immune system, such as erythrocytes, monocytes, and lymphocytes. Significantly less is known regarding the function of NRs in regulating the fate of hematopoietic stem cells (HSCs), the self-renewing, pluripotent cells that give rise to the entirety of the blood and immune systems throughout the lifetime of an individual. Several recent studies suggest, either directl… Show more

“…Similarly, systemic treatment of mice with the pan RAR inverse agonist BMS493 reduced pre-μDC, whereas intraperitoneal administration of all trans-RA or the RARα agonist AM580 increased the frequency and number of pre-μDCs dramatically. RA regulates hematopoietic progenitor development, and modulation of RARa/RXR signaling regulates hematopoietic stem cell (HSC) differentiation, proliferation, and self-renewal 1921 . Furthermore, RA modulates neutrophil differentiation from myeloid progenitors under steady state conditions 22 .…”

“…NR4A2, also known as Nurr1, is an orphan nuclear receptor [19][22], and its function in dopaminergic neuron signalling has been widely known. Increasing evidence suggests the role of NR4A2 in inflammatory responses during arthritis and psoriasis [23], [24], and NR4A2 may also serve as a regulatory element for reducing immune-mediated tissue damage [25].…”

“…
Thyroid hormone stimulates erythropoiesis and clinical hypothyroidism is often complicated by reductions in red cell mass and total blood volume, and by normocytic normochromic anemia [1][2][3][4]. Clinically, bone marrow has been observed to show erythroid hyperplasia in patients with hyperthyroidism [5].
