This study was planned to determine the prevalences of the Nosema spp. and the black queen-cell virus (BQCV) among honeybees (Apis mellifera) raised in the province of Van by PCR and to determine the molecular characteristics of the determined isolates. A total of 260 adult worker bees from 26 colonies at 5 apiary locations belonging to the province of Van in April and May 2015 were collected for this reason. Samples were examined microscopically. In the case of positivity, spore identification was done by multiplex PCR. Reverse transcription/PCR analysis (RT/PCR) was carried out for the BQCV analysis. At the end of the microscopic examination, Nosema spp. spores were detected in 8 out of 26 colonies (32.5%). The result of multiplex-PCR revealed Nosema ceranae positivity in all of the samples, but no Nosema apis was determined. As a result of the RT/PCR tests of the samples BQCV was detected in 23 (88.5%) of the total 26 colonies. This study is the first to investigate Nosema spp. and BQCV with the PCR technique in bees raised in the province of Van.

Honey bees (Apis mellifera L.) are the most effective pollinators for different crops and wild flowering plants, thus maintaining numerous ecosystems in the world. However, honey bee colonies often suffer from stress or even death due to various pests and diseases. Among the latter, nosemosis is considered to be one of the most common diseases, causing serious damage to beekeeping every year. Here, we present, for the first time, the effects from the application of the herbal supplements NOZEMAT HERB® (NH) and NOZEMAT HERB PLUS® (NHP) for treating N. ceranae infection and positively influencing the general development of honey bee colonies. To achieve this, in autumn 2019, 45 colonies were selected based on the presence of N. ceranae infections. The treatment was carried out for 11 months (August 2019–June 2020). All colonies were sampled pre- and post-treatment for the presence of N. ceranae by means of light microscopy and PCR analysis. The honey bee colonies’ performance and health were evaluated pre- and post-treatment. The obtained results have shown that both supplements have exhibited statistically significant biological activity against N. ceranae in infected apiaries. Considerable enhancement in the strength of honey bee colonies and the amount of sealed workers was observed just one month after the application of NH and NHP. Although the mechanisms of action of NH and NHP against N. ceranae infection are yet to be completely elucidated, our results suggest a new holistic approach as an alternative therapy to control nosemosis and to improve honey bee colonies’ performance and health.

Nosemosis is known as a serious disease of adult honey bees, Apis mellifera L. (Hymenoptera: Apidae) caused by Nosema apis and N. ceranae which are obligate intracellular microsporidian parasites. The parasites infect the epithelial cells of honey bee ventriculum and lead to critical changes in midgut mucosa that cause digestive and metabolic disorders. Accordingly, the infestation causes the death of adult honey bees and leads to great economic losses for beekeeping industry worldwide. Seasonal patterns of nosemosis are consistent and mostly observed in the spring or autumn months, with the highest spore counts and viability. The aim of this work was to evaluate the seasonality of nosemosis in the light of previous literature and specifically investigate the presence of Nosema spp. during the summer season including June, July, and August from various locations in Ankara. Honey bee individuals were collected from 80 apiaries located in 14 different areas in Ankara. The samples were analyzed from pools of ten adult honey bees per population using digestion methods. Before analysis, the anesthesia was induced by cold (30 sec, -80 ℃) on the bees. According to obtained data, 12 out of 80 (15%) sampled apiaries were infected with Nosema spp. spores. Infected apiaries were mainly located in the central and north parts of Ankara. Results show nosemosis might be detrimental to honey bee colonies and its productivity in the summer months. Therefore, the treatment might be needed when infections of Nosema spp. reach to an infectious level even in summer.
