microRNAs in the Regulation of Melanogenesis

Abstract: Melanogenesis is the process leading to the synthesis of melanin, the main substance that influences skin color and plays a pivotal role against UV damage. Altered melanogenesis is observed in several pigmentation disorders. Melanogenesis occurs in specialized cells called melanocytes, physically and functionally related by means of autocrine and paracrine interplay to other skin cell types. Several external and internal factors control melanin biosynthesis and operate through different intracellular signaling… Show more

“…Recently, it has been appreciated how EVs secreted into interstitial spaces mediate the interchange between keratinocytes and melanocytes. Keratinocytes communicate with melanocytes via sEVs carrying miRNAs such as miR-203 and miR-3196 with the capacity to modulate pigmentation in addition to known molecules such as the α-melanocyte stimulating hormone (α-MSH) and the adrenocorticotropic hormone (ACTH) ( Hushcha et al, 2021 ). These keratinocytes-derived vesicles increase tyrosinase activity, pigmentation genes and melanin content in recipient melanocytes to stimulate more melanosome production ( Lo Cicero et al, 2015b ).…”

“…Melanogenesis can be regulated by epigenetic factors, such as miRNAs, DNA methylation, and posttranscriptional controllers [140,141]. Lower expression of miR-675 and miR-1299 has been demonstrated in skin with melasma compared with the adjacent area [142,143].…”

“…As we all know, tyrosinase plays an important role in the process of melanogenesis (Hushcha et al, 2021). Tyrosine can be catalyzed, oxidized and polymerized to be pheomelanin.…”
