microRNA strand selection: Unwinding the rules

Abstract: microRNAs (miRNAs) play a central role in the regulation of gene expression by targeting specific mRNAs for degradation or translational repression. Each miRNA is post‐transcriptionally processed into a duplex comprising two strands. One of the two miRNA strands is selectively loaded into an Argonaute protein to form the miRNA‐Induced Silencing Complex (miRISC) in a process referred to as miRNA strand selection. The other strand is ejected from the complex and is subject to degradation. The target gene specifi… Show more

“…An miRNA gene cluster is composed of two or more related miRNA genes that may target specific mRNAs in the regulatory network. An miRNA duplex contains two “arms,” and the dominant one is the one which usually forms the mature miRNA; the other usually tends to be degraded, and is known as miRNA ∗ ( Medley et al, 2021 ). In recent years, studies have found that miRNAs and miRNAs ∗ can co-exist under certain circumstances ( Correia de Sousa et al, 2019 ; Dong et al, 2019 ; Zhang Z. et al, 2019 ).…”

“…Argonaute protein (Ago) determines which strand of a miRNA duplex will serve as a silencing guide strand and supposedly favors the one whose 5' sequence (the first four nucleotides) comprises less thermodynamically stable end of the duplex and whose first nucleotide is preferentially U (>A>>C and G). However, only 25% of human miRNA are chosen accordingly to both conditions and as much as 25% totally eludes these rules (Medley, Panzade, & Zinovyeva, 2020). Likewise, the strand of shRNA-derived siRNA designed to be a passenger strand can also be loaded into Ago (Sheng et al, 2020), thus doubling the number of potential off-targets.…”

“…In addition, miRNA biogenesis is controlled posttranscriptionally on multiple levels, and a number of cis‐ and trans‐acting factors modulate this process (Ha & Kim, 2014; Michlewski & Cáceres, 2019). Improper amiRNA design may result in inefficient processing, the generation of siRNA variants with seed sequence changes or the induction of arm switching—a process that leads to the release of the passenger strand of siRNA (Medley et al, 2020).…”
